Self-assembly of 3D interiors and iridescence properties of poly(β-hydroxybutyric acid-co-β-hydroxyvaleric acid) (PHBV) periodic crystals are examined using microcopy techniques and microbeam X-ray diffraction. Morphology of PHBV can be tailored by crystallizing in presence of poly(vinyl acetate) (PVAc) or poly(trimethylene adipate) (PTA) for displaying desired periodicity patterns. The regular alternate-layered lamellae of banded PHBV crystal aggregates, resembling the structures the natural mineral moonstone or nacre, are examined to elaborate the origin of light interference and formation mechanisms of periodic lamellar aggregation of PHBV spherulites.

Two new diterpenoids, pachydictyol B (1a/1b) and C (2), were isolated from the dichloromethane extract of the marine brown alga, Dictyota dichotoma, collected from the Red Sea coast of Egypt, along with the known metabolites, pachydictyol A (3a), dictyol E (4), cis-africanan-1α-ol (5a), fucosterol (6), tetrahydrothiophen-1,1-dioxide and poly-β-hydroxybutyric acid. GC-MS analysis of the nonpolar fractions also indicated the presence of β-bourbonene and nonanal, along with three hydrocarbons and five fatty acids or their simple derivatives, respectively. Some particular cell components of intact Bacillus cereus could be detected and identified by Fourier transform infrared spectroscopy (FTIR). FTIR were collected directly, then the FTIR spectra of the samples were converted into second derivative spectra. Typical marker bands were used to identify these bacterial cell components such as capsules, endospores or storage materials.

Microorganisms rarely occur as individual cells in nature and are, instead, organized in complex multicellular communities such as colonies, fruiting bodies, or biofilms. Interest in the natural microbial life-style has increased during the last decade and a whole plethora of techniques has been used to gain insight into the development, structure and composition of diverse microbial communities. We have developed a technique for investigating the spatial heterogeneity of microbial growth in macro-colonies which essentially entails excision of the colonies with the underlying agar, freezing and subsequent cryotoming of the colonies, then FTIR microspectroscopic mapping of the cryosections.
View Article and Find Full Text PDFFrom genomic libraries of Alcaligenes eutrophus H16 in lambda L47 and in pVK100, we cloned DNA fragments which restored the wild-type phenotype to poly(beta-hydroxybutyric acid) (PHB)-leaky mutants derived from strains H16 and JMP222. The nucleotide sequence analysis of a 4.5-kb region of one of these fragments revealed two adjacent open reading frames (ORF) which are relevant for the expression of the PHB-leaky phenotype.
