The conformation of oxytocin, the neurohypophyseal nonapeptide hormone, in solution in deuterated dimethyl sulfoxide has been determined by 1H-nmr. The structural determination is based on the experimental data set of nuclear Overhauser effect restraints. Obtained after the restrained molecular dynamics simulation on an initial structure of extended conformation, five resultant structures satisfy the experimental restraints well.
View Article and Find Full Text PDFThis paper describes the synthesis and biological activity of [5-L-asparagine-alpha,beta,beta-d(3)]-deamino-oxytocin. The model peptide S-benzyl-beta-mercaptopropionyl-L- asparaginyl-alpha,beta,beta-d(3)-glycinamide was also prepared to observe the effect of the conditions of peptide synthesis on the deuterium content. It was found that normal synthetic procedures could be used without a significant loss in the deuterium content.

View Article and Find Full Text PDFX-ray studies of crystalline deamino-oxytocin (1-beta-mercaptopropionic acid-oxytocin) have established the existence of both wet and dry forms. A remarkable degree of similarity exists between the two forms even though there are changes in cell dimensions and space group on drying. The dry form of 1-gamma-mercaptobutyric acid-oxytocin is very similar to that of deamino-oxytocin.
