Line-Shifting Triarylmethyl Radicals for Imaging of Enzyme Activity Using Overhauser-Enhanced Magnetic Resonance Imaging: Application to Alkaline Phosphatase.

Chem Biomed Imaging

January 2025

In Vivo Multifunctional Magnetic Resonance Center, Robert C. Byrd Health Sciences Center, West Virginia University, Morgantown, West Virginia 26506, United States.

Enzyme catalytic activities are critical biomarkers of tissue states under physiological and pathophysiological conditions. However, the direct measurement and imaging of enzyme activity remains extremely challenging. We report the synthesis and characterization of the first stable triarylmethyl (TAM) radical substrate of alkaline phosphatase (TAM-ALPs).

Methylmalonic acid (MMA) is a small molecule produced during the metabolism of propionate and branched-chain amino acids. Recently, it has been reported that the blood concentration of MMA increases with age and promotes lung cancer metastasis. However, little is known regarding its effects on cancers other than lung cancer.

Isolation of Natural Products Using Preparative TLC.

Methods Mol Biol

January 2025

Department of Biomedical and Pharmaceutical Sciences, Graduate School, Kyung Hee University, Seoul, Republic of Korea.

Known for their diverse and potent physiological activities, natural products continue to be essential for the discovery and development of new drugs. This chapter explores the pivotal role of preparative thin-layer chromatography (Prep-TLC) in the isolation of natural products. This chapter begins with an understanding of the historical significance and structural complexity of natural products, and discusses the problems caused by complex mixtures present in extracts, as well as the multifunctionality, cost-effectiveness, and compatibility with different sample types of Prep-TLC to address these challenges.

Regulation of circadian gene activity in fibroblasts from ADHD patients through Rosiglitazone: a pilot study.

J Neural Transm (Vienna)

January 2025

Chair of Vascular Neurology, Dementia and Ageing, University Hospital Essen, Essen University Medical School, University of Duisburg-Essen, 45147, Essen, Germany.

Attention-deficit/hyperactivity disorder (ADHD) is a frequently observed condition, with about 70% of individuals diagnosed with ADHD experiencing irregular sleep-wake patterns. Beyond the primary symptoms of ADHD, there is a significant overlap with sleep-related issues, indicating that disrupted sleep patterns may exacerbate ADHD symptoms. ADHD-related sleep problems can be traced to a delayed circadian rhythm and a later onset of melatonin production.

Introduction: Threats to our survival are often posed by the environment in which humans have evolved or live today. Animal and human ancestors developed complex physiological and behavioral response systems to cope with two types of threats: immediate physical harm from predators or conspecifics, triggering fear, and the risk of infections from parasites and pathogens leading to the evolution of the behavioral immune system (BIS) with disgust as the key emotion. Here we ask whether the BIS has adapted to protect us from pandemic risks or poisoning by modern toxic substances.
