Analysis of mice carrying mutant T-cell antigen receptor (TCR) genes indicates that TCR-beta gene rearrangement or expression is critical for the differentiation of CD4-CD8- thymocytes to CD4+CD8+ thymocytes, as well as for the expansion of the pool of CD4+CD8+ cells. TCR-alpha is irrelevant in these developmental processes. The development of gamma delta T cells does not depend on either TCR-alpha or TCR-beta.

Chimeric antigen receptor (CAR) T-cell therapy has led to significant advances in the treatment of blood cancers such as leukemia, lymphoma, and multiple myeloma, and now shows promise for solid tumors. This type of immunotherapy can achieve high response rates in patients with hematologic malignancies, but carries serious adverse reactions, including cytokine release syndrome and immune-effector cell-associated neurotoxicity syndrome. This article describes CAR T-cell therapy, guidance for primary care providers caring for patients undergoing therapy, and the ongoing need for research to enhance CAR T-cell therapy's safety and effectiveness.

T cells recognize a wide range of pathogens using surface receptors that interact directly with peptides presented on major histocompatibility complexes (MHC) encoded by the HLA loci in humans. Understanding the association between T cell receptors (TCR) and HLA alleles is an important step towards predicting TCR-antigen specificity from sequences. Here we analyze the TCR alpha and beta repertoires of large cohorts of HLA-typed donors to systematically infer such associations, by looking for overrepresentation of TCRs in individuals with a common allele.

Human papillomavirus 16 and human papillomavirus 18 have been associated with different life-threatening cancers, including cervical, lung, penal, vulval, vaginal, anal, and oropharyngeal cancers, while cervical cancer is the most prominent one. Several research studies have suggested that the oncoproteins E6 and E7 are the leading cause of cancers associated with the human papillomavirus infection.
