Background: Inflammation and subsequent mitochondrial dysfunction and cell death worsen outcomes after revascularization in ischemic stroke. Receptor-interacting protein kinase 1 (RIPK1) activated dynamin-related protein 1 (DRP1) in a NLRPyrin domain containing 3 (NLRP3) inflammasome-dependent fashion and Hypoxia-Inducible Factor (HIF)-1α play key roles in the process. This study determined how phenothiazine drugs (chlorpromazine and promethazine (C + P)) with the hypothermic and normothermic modality impacts the RIPK1/RIPK3-DRP1 and HIF-1α pathways in providing neuroprotection.

Chlorpromazine (CPZ), a first-generation antipsychotic, is widely used in treating schizophrenia and other psychiatric disorders. However, CPZ is also associated with an increased likelihood of sudden cardiac death, and the underlying mechanisms remain unclear. In our study, we aimed to determine the CPZ-induced changes in some members of the heat shock protein family in rat hearts and further explore the possible mechanisms of CPZ-induced cardiotoxicity.

Decapod crustaceans (crabs, hermit crabs, lobsters, crayfish, shrimps, prawns) are sentient beings, not only responding to noxious stimuli but also being capable of feeling pain, discomfort, and distress. General anaesthesia aims at producing analgesia, immobilization, and unconsciousness, while sedation reduces consciousness, stress, and anxiety, though without analgesia. Anaesthesia is recommended to ensure animal welfare and suppress nociception, pain, and suffering in painful and distressing practice that impairs decapods' welfare.
