Leptospirosis and dengue share similar unspecific symptoms, complicating differential diagnosis in endemic regions. This overlap is further exacerbated by the often-underrecognized nature of leptospirosis, resulting in low clinical suspicion among healthcare providers. Understanding the scale of underdiagnosed leptospirosis cases, particularly among dengue-negative patients, is critical for improving public health responses.

Leptospirosis is a widespread zoonotic disease that causes severe health complications with no approved vaccine which provide broad range protection. Mice are slowly being accepted as alternative models for investigation of leptospiral infection. The strain most used to analyse sublethal disease (C3H-HeJ) expresses a gene in its immune cells that is hyporesponsive to leptospiral LPS and thus the model is deemed immunocompromised. To help resolve this valid scientific concern we did a study in which we compared infection with serovar Copenhageni Fiocruz in mice expressing fully competent (C3H-HeN, C57BL6) versus hyporesponsive mice (C3H-HeJ) over a period of two weeks.

Leptospirosis is a major cause of acute febrile illness, often presenting with non-specific symptoms that can lead to misdiagnosis. Early laboratory diagnosis is essential for confirmation to avoid misdiagnosis and ensure appropriate management. This study aimed to identify and produce a recombinant protein, approximately 25 kDa, with high antigenicity for diagnostic applications.
