Assessing plasmatic transport inhibitors of thyroid hormone in mammals in the Xenopus Eleutheroembryonic Thyroid Assay (XETA).

Environ Sci Pollut Res Int

November 2024

BASF SE, Agricultural Solutions - Ecotoxicology, Speyerer Strasse 2, 67117, Limburgerhof, Germany.

The Xenopus Eleutheroembryonic Thyroid Assay (XETA, OECD TG 248) was established as an alternative to the Amphibian Metamorphosis Assay (AMA, OECD TG 231) for the analysis of (anti-)thyroid activity of chemicals. The XETA is a New Approach Method (NAM) since the embryonic life stages used in the assay are not yet feeding independently, which renders the assay to be considered a non-animal test under many national laws. Physiologically, the used embryos are not fully developed yet, and thus there are limitations to the XETA for detecting certain mechanisms along the hypothalamic-pituitary-thyroid (HPT) axis.

Article Synopsis
  • The study aimed to investigate how the hypothalamic-pituitary-thyroid (HPT) axis responds to thyroid hormones in pregnant women, specifically focusing on the relationship between central sensitivity to these hormones (measured by the Thyroid Feedback Quantile-based Index or TFQI) and urinary iodine (UI) levels.
  • A total of 1,102 blood and urine samples were collected from healthy pregnant women, assessing levels of TSH, fT4, anti-thyroid antibodies, and UI across different trimesters and postpartum.
  • Results showed a negative correlation between TFQI and UI levels, indicating higher central sensitivity to thyroid hormones when UI is lower; interestingly, this relationship varied depending on the trimester and them

Hazard assessment of fenozan, a released non-intentionally added substance from polyester-based can coating.

Food Addit Contam Part A Chem Anal Control Expo Risk Assess

December 2024

CTM UMR1231, Nutrition Physiology and Toxicology Team (NUTox), Dijon, France.

Since the safety of new-generation polyester-based internal coatings regarding the migration of non-intentionally added substances (NIAS) is poorly documented, studies are needed to identify NIAS originating from these food-contact materials (FCM). The aim of this study was to identify volatile and semi-volatile NIAS from polyester-based coatings in order to assess their hazard and ensure consumers' safety with regard to exposure from canned food. Extraction and migration tests were carried out on a single polyester-coated tin plate (5 batches) using two solvents: acetonitrile and ethanol 95%, then FCM's extracts and migrates were analysed by GC-MS.

Background: Chronic spontaneous urticaria (CSU) is a highly prevalent and difficult to manage cutaneous disease characterized by the presence of recurrent urticaria, angioedema, or both, for a period of 6 weeks or longer. One of the biological treatments used for patients with CSU with an autoimmune background and bad control of the disease is omalizumab, an anti-IgE monoclonal antibody. The understanding of the mechanism of action of this biological drug in CSU along with the identification of potential biomarkers of clinical response can be helpful in the personalized management of the disease.

Thyroid Hormone and Diabetes Mellitus Interplay: Making Management of Comorbid Disorders Complicated.

Horm Metab Res

December 2024

Department of Pharmacology, Institute of Pharmacy, Nirma University, Ahmedabad, India.

Insulin and thyroid hormones play important roles in our body. Insulin helps regulate the glucose level while the thyroid hormones affect various cells and tissues, metabolizing protein, lipids, and glucose. Hyperthyroidism and thyrotoxicosis are potential hazards for type 2 diabetes mellitus.
