The widespread adoption of chimeric antigen receptor (CAR) T-cell therapy has been limited by complex, resource-intensive manufacturing processes. This review discusses the latest innovations aiming to improve and streamline CAR T-cell production across key steps like T-cell activation, genetic modification, expansion, and scaling. Promising techniques highlighted include generating CAR T cells from non-activated lymphocytes to retain a stem-like phenotype and function, non-viral gene transfer leveraging platforms like transposon and CRISPR, all-in-one fully automated bioreactors like the CliniMACS Prodigy and the Lonza Cocoon, rapid CAR T-cell manufacturing via abbreviating or eliminating ex vivo T-cell culture, implementing decentralized point-of-care automated manufacturing platforms, and optimizing centralized bioreactor infrastructure integrating end-to-end automation.

Objectives: Due to the severe shortage of donor corneas for transplantation in China, corneal component transplantation may expand the available donor pool. This study aims to evaluate the safety and feasibility of corneal component transplantation by examining the distribution of hepatitis B surface antigen (HBsAg) in corneas from HBsAg-seropositive donors under different storage media.
Methods: Ten corneas (from 6 donors) donated between December 2019 and March 2021 and stored at the Eye Bank of Xiangya Third Hospital, Central South University, were analyzed.

The CD2-depleting drug alefacept (LFA3-Ig) preserved beta cell function in new-onset type 1 diabetes (T1D) patients. The most promising biomarkers of response were late expansion of exhausted CD8 T cells and rare baseline inflammatory islet-reactive CD4 T cells, neither of which can be used to measure responses to drug in the weeks after treatment. Thus, we investigated whether early changes in T cell immunophenotypes could serve as biomarkers of drug activity.

Anaplasmosis is a tick-borne disease (TBDs) caused by Anaplasma spp. In areas where TBDs are endemic, it is crucial to consider the animals' immunological status in relation to these diseases. The true prevalence of bovine anaplasmosis, the percentage of animals with protective antibodies against this TBD, and the diagnostic characteristics of three tests (multiplex polymerase chain reaction (mPCR), competitive-inhibition enzyme-linked immunosorbent assay (cELISA), and blood smear (BS)) were estimated using a Bayesian approach.
