In dogs maintained on low protein diets and subjected to phlebotomy over a long period of time, the inhalation of chloroform, for 30 minutes, produced uniform fatality within 48 hours. The histological changes of massive hepato-cellular destruction were observed at autopsy. Homocystine, in the amount of 2.0 gm., given orally 2 hours after or 2 hours prior to the administration of chloroform, protected dogs against the lethal action of the toxin. Inconstant changes in fibrinogen and icteric indices were observed in the protected animals, indicating some mild liver damage, but this was not correlated with the length of the previous depletion period, the phase of the experiment, or any other factor. No evidence was obtained that methyl groups are necessary for the protection of the liver by homocystine.

Background: Acute ischemic stroke (AIS) is one of the most common cerebrovascular diseases which accompanied by a disruption of aminothiols homeostasis. To explore the relationship of aminothiols with neurologic impairment severity, we investigated four aminothiols, homocysteine (Hcy), cysteine (Cys), cysteinylglycine (CG) and glutathione (GSH) in plasma and its influence on ischemic stroke severity in AIS patients.

Increased plasma concentrations of total homocysteine (tHcy; mild-moderate hyperhomocysteinemia: 15-50 μ tHcy) are considered an independent risk factor for the onset/progression of various diseases, but it is not known about how the increase in tHcy causes pathological conditions. Reduced homocysteine (HSH ∼1% of tHcy) is presumed to be toxic, unlike homocystine (∼9%) and mixed disulfide between homocysteine and albumin (HSS-ALB; homocysteine [Hcy]-albumin mixed disulfide, ∼90%). This and other notions make it difficult to explain the pathogenicity of Hcy because: (i) lowering tHcy does not improve pathological outcomes; (ii) damage due to HSH usually emerges at supraphysiological doses; and (iii) it is not known why tiny increments in plasma concentrations of HSH can be pathological.
