Cultured organotypic hippocampal slices (hOTCs) have become increasingly popular as a model for studying brain function. This model offers significant advantages over traditional in vitro methods, as they allow the examination of mid to long-term manipulations while preserving the structure of the dentate gyrus (DG) in the hippocampus. In this chapter, we focus on a protocol based on hOTCs of mouse entorhinal cortex and hippocampus, which by integrating techniques such as retroviral injections, immunohistochemistry, and microscopy imaging, physiological or pathological processes can be easily investigated.

This research introduces innovative tools for studying embryonic neurogenesis in the gecko Paroedura picta. Traditional research methods have been adapted for the gecko's unique biology, including variations of birthdating techniques and the implementation of EdU for tracking neuron generation. We also employ hybridization chain reaction (HCR) to detect specific mRNAs without the need for species-specific antibodies, providing a powerful and flexible tool for studying gene expression patterns in the gecko embryonic brain.

Background: Circadian rhythm disorders and NF-κB are closely linked and can exacerbate periodontitis. However, the mechanisms via which circadian rhythm-related genes influence periodontitis are not yet fully understood.
Objective: We investigated the effect of brain and muscle Arnt-like protein-1 (BMAL1) on the NF-κB pathway and downstream inflammatory factors on periodontitis.

Immunostaining Drosophila melanogaster brains is essential for exploring the mechanisms behind complex behaviors, neural circuits, and protein expression patterns. Traditional methods often involve challenges such as performing complex dissection, maintaining tissue integrity, and visualizing specific expression patterns during high-resolution imaging. We present an optimized protocol that combines cryosectioning with fluorescence staining and immunostaining.

Background: Brain tumors account for 2%-3% of all malignant neoplasms and 85%-90% of all primary tumors of the central nervous system with the 5-year survival rate of 35%. Additional biomarkers could help refine the molecular profile of brain tumors and prognosis of the disease.
Aim: To study differential expression patterns of the MRPS18 family genes in tumor tissue and the peripheral blood of patients with brain tumors of various types.
