Article Synopsis
  • * A newly designed FUCL probe, PT-1, shows high sensitivity and selectivity in detecting HSO, with a detection limit significantly lower than traditional methods (43 nM vs. 93 nM).
  • * PT-1 not only targets mitochondria for real-time monitoring of HSO in living cells but also successfully tracks HSO levels in mice experiencing drug-induced liver injury using FUCL imaging.

The early diagnosis of tumorigenesis is crucial for clinical treatment, but the resolution and sensitivity of conventional short-wavelength biomarkers are not ideal because of the complicated interference in living tissue. Herein, a nicotinamide adenine dinucleotide (NAD)-responsive probe with deep-red emissive ratiometric fluorescence was synthetized as a promising target for energy metabolism patterns during tumorigenesis. Interestingly, the solvents HPO and 2,2'-dithiodibenzoic acid enhanced the red emission (640 and 680 nm) of o-phenylenediamine-based carbon dots (CDs), leading to the formation of a nanoscale graphite-like skeleton covered with -P=O, -CONH-, -COOH and -NH on their surfaces.

Background: Choroideremia is a monogenic inherited retinal dystrophy that manifests in males with night blindness, progressive loss of peripheral vision, and ultimately profound sight loss, commonly by middle age. It is caused by genetic defects of the gene, which result in a deficiency in Rab-escort protein-1, a key element for intracellular trafficking of vesicles, including those carrying melanin. As choroideremia primarily affects the retinal pigment epithelium, fundus autofluorescence, which focuses on the fluorescent properties of pigments within the retina, is an established imaging modality used for the assessment and monitoring of affected patients.

The fabrication of self-reporting photosensitizers (PSs), enabling real-time evaluation of the extent of elimination of cancer cells, holds significant scientific importance in the photodynamic therapy (PDT) process. To address the intrinsic challenge of the short-wavelength light source, this work proposed an innovative approach of rational design second near-infrared (NIR-II, 1000-1700 nm) light-excited fluorescent PS systems (named , , and , respectively) through modulating aggregation and deaggregation based on assembling strategy. Therein, the suitable interplanar distance of adjacent linked with C-H···π interactions was an idea for relieving compact π···π packing for fluorescent imaging as well as elevating the spin-orbit coupling for reactive oxygen species (ROS) generation.

Inflammation is a complex physiological response involving various cellular and molecular events. Sulfur dioxide (SO), which is usually in the form of HSO and SO under physiological conditions, plays a crucial role in the regulation of inflammation and diseases. Frequency upconversion luminescence (FUCL) can realize the unique anti-Stokes process of long-wavelength excitation to short-wavelength emission; thus, it is a highly promising optical method for imaging due to its deep tissue penetration, low photo-damage, .
