Objective: To study whether quantitative analysis of M. tuberculosis mRNA could be used to assess bacterial viability.

Methods: The levels of M. tuberculosis mRNA were compared 24, 48, and 72 hours after M. tuberculosis H(37)R(V) was treated with no drug, 1.0 micro g/ml and 10.0 micro g/ml rifampicin, 0.2 micro g/ml and 1.0 micro g/ml isoniazid, 2 micro g/ml and 4 micro g/ml ethambutol, 2 micro g/ml and 4 micro g/ml streptomycin, 1.25 micro g/ml and 5.00 micro g/ml ofloxacin. The levels of mRNA were determined by quantitative PCR.

Results: After exposure to isoniazid, rifampicin, and streptomycin for 24 h, the level of M. tuberculosis H(37)R(V) was reduced markedly. Similarly, after exposure to isoniazid, streptomycin, ethambutol, and ofloxacin for 72 h, the level of M. tuberculosis H(37)R(V) was identical, but higher than that after exposure to rifampicin. Exposure of M. tuberculosis H(37)R(V) to rifampicin for 24 h reduced the levels of 85B mRNA to 0.02% of the control; exposure to other drugs for 24 h reduced the levels to 1% approximately 10% of the control, and for 72 h to less than 1% of the control.

Conclusions: The levels of mRNA can show the difference between M. tuberculosis H(37)R(V) exposed and unexposed to drugs, and are consistent with colony forming unit. mRNA could be used as a marker for assessing the viability of M. tuberculosis.

Background/objectives: The alarming rise in antibiotic resistance necessitates the discovery of novel antimicrobial agents. This study aims to design, synthesize, and evaluate new benzofuran-pyrazole-based compounds for their antimicrobial, antioxidant, and anti-inflammatory properties.

Methods: New benzofuran-pyrazole hybrid molecules were synthesized using the Vilsmeier-Haach reaction and other chemical processes.

Constructing two bifunctional tooth-targeting antimicrobial peptides for caries management: an in vitro study.

Clin Oral Investig

December 2024

Department of Stomatology, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology, No. 1095 Jiefang Avenue, Wuhan, Hubei Province, 430030, China.

Objectives: Caries is a significant public health challenge. Herein, novel tooth-targeting antimicrobial peptides (HABPs@AMPs) were developed by combining the antimicrobial peptide DJK-5 with hydroxyapatite (HA) binding peptides, providing a potential new strategy for caries management.

Materials And Methods: The minimal inhibitory concentration (MIC) and minimal biofilm inhibitory concentration (MBIC) values of HABPs@AMPs were determined via micro-broth dilution and crystal violet staining.

Article Synopsis
  • - This study investigates the health benefits of ripe Meyna spinosa fruit extracts, focusing on their abilities to act as antioxidants, antiproliferatives, and anti-inflammatories, supported by chemical analysis identifying key bioactive compounds.
  • - The ethyl acetate extract showed the highest antioxidant activity, while both ethyl acetate and methanol extracts reduced cancer cell growth significantly and demonstrated strong anti-inflammatory effects by decreasing levels of various inflammatory markers in immune cells.
  • - In animal models, the ethyl acetate extract effectively reduced swelling from carrageenan-induced inflammation by 23.45%, suggesting potential therapeutic applications for inflammation and cancer treatment.
