Background And Aim: Cannabis use disorder (CUD) is strongly influenced by genetic factors; however the mechanisms underpinning this association are not well understood. This study investigated whether a polygenic risk score (PRS) based on a genome-wide association study for CUD in adults predicts cannabis use in adolescents and whether the association can be explained by inter-individual variation in structural properties of brain white matter or risk-taking behaviors.

Design And Setting: Longitudinal and cross-sectional analyses using data from the IMAGEN cohort, a European longitudinal study integrating genetic, neuroimaging and behavioral measures.

For the last 38 years, all neuroprotective agents for patients with ischemic stroke have failed in clinical trials. The innate immune system, particularly microglia, is a much-discussed target for neuroprotective agents. Promising results for neuroprotection by inhibition of integrins with drugs such as natalizumab in animal stroke models have not been translated into clinical practice.

Alzheimers Dement

December 2024

Department of Molecular Imaging & Therapy, Austin Health, Heidelberg, VIC, Australia.

Background: Tau PET is instrumental in tracking the longitudinal progression of Alzheimer's disease (AD). F-MK6240 is a high affinity tracer targeting the 3R/4R paired helical filaments of tau in AD. We aimed to evaluate the early phase of the natural progression of tau accumulation using F-MK6240.

Background: Normative models (NM) of brain metrics based on large, diverse populations offer novel strategies to detect individual brain abnormalities. To create an age-dependent statistical model of brain microstructure over the human lifespan, we built the largest multi-site NM of white matter (WM) diffusion tensor imaging (DTI) metrics based on 54,591 subjects. We used state-of-the-art tools to adjust for site-dependent effects.

Alzheimers Dement

Imaging Genetics Center, Mark and Mary Stevens Neuroimaging & Informatics Institute, University of Southern California, Marina del Rey, CA, USA.

Background: High body mass index (BMI) is a risk factor for dementia, and prior diffusion-weighted magnetic resonance (dMRI) work has shown that higher BMI is associated with lower white matter (WM) integrity. The tensor distribution function (TDF) is an advanced dMRI model that is sensitive to the effects of both healthy aging and Alzheimer's disease (Nir et al., 2017; Lawrence et al.
