Published: August 2003

Objective: To investigate the chromatin status of oocytes and their ploidity in order to consider possible genetic reasons for their unsuccessful fertilization in vitro.

Study Design: Eighty-six unfertilized in vitro human oocytes were scored in six groups according to their chromatin status. Analysis was performed by cytogenetics and by fluorescent in situ hybridization (FISH).

Results: 73.26% of oocytes were successfully analyzed: 44.19% by classical cytogenetics and 17.44% by FISH. 11.63% of the analyzed cells contained sperm chromosomes with premature chromosome condensation (PCC). Among karyotyped oocytes, a high aneuploidy rate was found (5.26% haploid abnormal, 21.05% hypohaploid, 36.84% hyperhaploid and 7.89% diploid). FISH for chromosomes X, 1, 5 and 19 performed on unsuitable for karyotyping metaphase plates revealed 13.33% aneuploid cells.

Conclusion: Analysis of fertilization failure could be more informative when a combination of methods is applied.

Objective: This study aimed to investigate whether NKAP (nuclear factor κB activating protein) serves as a prognostic marker and predictive biomarker for immunotherapy response in head and neck squamous cell carcinoma (HNSCC).

Methods: A retrospective cohort study combined with in vitro analyses was conducted. NKAP mRNA expression levels were assessed in 520 HNSCC tumor tissues and 44 normal tissues from the TCGA dataset and validated in a clinical cohort (n=32).

Objectives: Nonalcoholic fatty liver disease (NAFLD) is known to disrupt testicular anti-oxidant capacity, leading to oxidative stress (OS) that can negatively affect male fertility by damaging sperm DNA. Heat shock proteins (HSP70 and HSP90), in association with transitional proteins (TP1 and TP2), play crucial roles in protecting sperm DNA integrity in oxidative conditions. Whiteleg shrimp protein hydrolysates (HPs) exhibit anti-oxidant properties, prompting this study to explore the potential of HPs in ameliorating NAFLD-induced testicular damage.

Isolation of proteins on chromatin (iPOC) reveals signaling pathway-dependent alterations in the DNA-bound proteome.

Signaling pathways often convergence on transcription factors (TFs) and other DNA-binding proteins (DBPs) that regulate chromatin structure and gene expression, thereby governing a broad range of essential cellular functions. However, the repertoire of DBPs is incompletely understood even for the best-characterized pathways. Here, we optimized a strategy for the isolation of Proteins on Chromatin (iPOC) exploiting tagged nucleoside analogues to label the DNA and capture associated proteins, thus enabling the comprehensive, sensitive, and unbiased characterization of the DNA-bound proteome.

Background: The newest NOX isoform, NOX5, has been found in mammalian spermatozoa. Many physiological and pathological situations in spermatozoa are mediated by reactive oxygen species (ROS). NOX5 is the main source of ROS in spermatozoa.

Merkel cell carcinoma (MCC) is an uncommon aggressive neoplasm, usually arising in sun-exposed skin of the head and neck. By immunohistochemistry, KRT20 and MCPyV positivity are found in about 90% and 80% of MCCs, respectively. Noteworthy, viral status in lip/oral cavity MCCs is poorly known.
