Gender-specific and parity-dependent acquired antibody recognition is characteristic of variant surface antigens (VSA) expressed by chondroitin sulfate A (CSA)-adherent Plasmodium falciparum involved in pregnancy-associated malaria (PAM). However, antibody recognition of recombinant products of a specific VSA gene (2O2var1) implicated in PAM and transcribed by a CSA-adhering parasite line did not have these characteristics. Furthermore, we could not demonstrate preferential transcription of 2O2var1 in the CSA-adhering line versus the unselected, parental isolate. Our data call for circumspection regarding the molecular identity of the parasite ligand mediating adhesion to CSA in PAM.

DNA-decorated hapten (DDH)-based immunoassays have emerged, demonstrating supreme advantages in sensing applications because of their excellent sensitivity, specificity, and reliability. DDH combines both a recognition element (hapten) and a signal transduction element (DNA portion) with its highly programmable DNA structure enabling the trigger of signal transduction following a recognition event, thereby introducing a novel signal transduction mechanism to immunoassays. In this review, we provide a critical overview of recent research in the DDH-based immunoassays, which are designed to detect specific small molecules and antibodies.

Background: Tau phosphorylated at threonine-217 (tau), which can be measured in plasma and CSF using antibodies, is one of the most promising early biomarkers for Alzheimer's disease (AD). Little was known, however, about the cellular and subcellular distributions of tau, how those levels change during disease progression, the factor(s) that provoke tau accumulation, or functional consequences of tau phosphorylation at T217. Immune checkpoint inhibitors (ICIs), such as pembrolizumab, have revolutionized cancer treatment by enhancing the immune system's response to malignancies. However, these therapies are associated with immune-related adverse events (irAEs), including neuromuscular complications such as myasthenia gravis, myositis, and myocarditis. We describe two male patients, aged 67 and 68, with small cell and non-small cell lung cancers, who developed progressive neuromuscular symptoms, including ptosis, diplopia, and generalized weakness, after receiving pembrolizumab.

We report here the use of antibody-DNA conjugates (Ab-DNA) to activate the collateral cleavage activity of the CRISPR-Cas12a enzyme. Our findings demonstrate that Ab-DNA conjugates effectively trigger the collateral cleavage activity of CRISPR-Cas12a, enabling the transduction of antibody-mediated recognition events into fluorescence outputs. We developed two different immunoassays using an Ab-DNA as activator of Cas12a: the CRISPR-based immunosensing assay (CIA) for detecting SARS-CoV-2 spike S protein, which shows superior sensitivity compared with the traditional enzyme-linked immunosorbent assay (ELISA), and the CRISPR-based immunomagnetic assay (CIMA).
