Published: June 2003

Objective: To study the effects of basic fibroblast growth factor (bFGF) on the praxiology and cerebral acetylcholinesterase (AchE) fiber density of kainic acid-lesioned rat models of Alzheimer disease (AD).

Methods: AD models were induced in 30 normal adult rats by damaging the rat nucleus basalis of Meynert (NBM) with kainic acid, and the models were then assigned into 3 groups to receive cerebroventricular infusion with bFGF, saline or nothing for treatment, serving respectively as the treatment group at 30 min, 1, 3 and 7 d after the injury, sham treatment group or injury group. Another 10 rats were used as control group, which received saline injections into the NBM without further treatment. The learning and memory abilities of the rats were measured through Y-maze test 30 d after the operations, and AchE cytochemical study was conducted to calculate the density of the AchE fibers in the hippocampus and forebrain of the rats.

Results: In comparison with the injury group, improvement was noted in the memory ability of rats with bFGF treatment and the density of AchE fiber was also significantly increased (P<0.01), but the improvement in both respects failed to reach the normal level (P<0.01).

Conclusions: AD model can be successfully established by damaging the NBM with kainic acid, and bFGF is beneficial in improving the impaired learning and memory abilities and increasing the density of AchE fibers in the basal forebrain cortex and hippocampus in the models.

Cardiac dysfunction and adverse consequences induced by cardiac fibrosis have been well documented. However, the cardiac fibrosis pathway in chronic heart failure (CHF) remains unclear, and it is therefore necessary to conduct further research for the sake of developing more effective therapeutic strategies for CHF. Some recent studies suggest that Pericarpium Trichosanthis (PT) may help improve the progression of fibrotic diseases.

This study evaluated the antioxidant and antiproliferative effects of aqueous, ethanolic and methanolic extracts of Sedum nicaeense flowers and leaves. The MTT assay assessed cytotoxicity against colorectal cancer cells (Caco-2, HCT-116), breast cancer cells (T47D, MCF-7) and normal fibroblasts (MRC-5), while the ferric-reducing antioxidant power (FRAP) assay measured antioxidant capacity. Essential oils from flowers and leaves were analyzed using gas chromatography-mass spectrometry (GC-MS).

IL-33, a neutrophil extracellular trap-related gene involved in the progression of diabetic kidney disease.

Inflamm Res

January 2025

Department of Nephrology, First Affiliated Hospital of Naval Medical University, Shanghai Changhai Hospital, Shanghai, China.

Background: Chronic inflammation is well recognized as a key factor related to renal function deterioration in patients with diabetic kidney disease (DKD). Neutrophil extracellular traps (NETs) play an important role in amplifying inflammation. With respect to NET-related genes, the aim of this study was to explore the mechanism of DKD progression and therefore identify potential intervention targets.

Potassium Current Signature of Neuronal/Glial Progenitors in Amniotic Fluid Stem Cells.

Cells

January 2025

Department of Chemistry, Biology and Biotechnologies, University of Perugia, Via dell'Elce di Sotto 8, 06123 Perugia, Italy.

Article Synopsis
  • Amniotic fluid contains stem cells (AF-SCs) that have potential uses in regenerative medicine for treating various injuries and diseases.
  • When exposed to basic Fibroblast Growth Factor (bFGF), AF-SCs show the ability to survive and migrate in a rat brain model, resembling characteristics of neuronal/glial progenitor cells.
  • The study employs electrophysiological techniques to identify specific potassium currents in AF-SCs and confirms that histamine can influence calcium dynamics and potassium current activation in these cells.

The Rat-1 cell line was established as a subclone of the parental rat fibroblastoid line F2408, derived from Fisher 344 rat embryos. Rat-1 cells are widely used in various research fields, especially in cancer biology, to study the effects of oncogenes on cell proliferation. They are also crucial for investigating signal transduction pathways and play a key role in drug testing and pharmacological studies due to their rapid proliferation.
