The one-dimensional double helicate, [NH4][Mo2O4Gd(H2O)6(L-C4H2O6)2] x 4H2O (1), which was synthesized by the reaction of GdCl3, L-tartaric acid and ammonium molybdate in acidified water solution, is built up by two heft-handed single-helical chains, linked up further by eight-coordinated GdIII pieces in an enantiopure left-handed double helical configuration, of which each helix is formed by L-tartrate bridged six-coordinated MoVI atoms.

Helical structures such as right-handed double helix for DNA and left-handed α-helix for proteins in biological systems are inherently chiral. Importantly, chirality at the nanoscopic level plays a vital role in their macroscopic chiral functionalities. In order to mimic the structures and functions of natural chiral nanoarchitectures, a variety of chiral nanostructures obtained from artificial helical polymers are prepared, which can be directly observed by atomic force microscopy (AFM), scanning tunneling microscopy (STM), scanning electron microscopy (SEM), and transmission electron microscopy (TEM).

This work reports single-crystal X-ray diffraction (XRD), Scanning Tunneling Microscopy (STM), and quantum mechanics calculations of the 3-helical peptide Z-(Aib)-L-Dap(Boc)-Aib-NHiPr (Aib, α-aminoisobutyric acid; Dap, 2,3-diaminopropionic acid; Z, benzyloxycarbonyl; Boc, t-butoxycarbonyl). The peptide forms a double-helical superstructure, studied by XRD and STM. Such architecture is rare in short peptides.

Until now, Z-form RNAs were believed to only adopt a left-handed double-helix structure. In this study, we describe the first observation of a right-handed Z-form RNA in NMR solution formed by L-nucleic acid RNA and present the first resolution of structure of the complex between a right-handed Z-form RNA and a curaxin ligand. These results provide a platform for the design of topology specific to Z-form-targeting compounds and are valuable for the development of new potent anticancer drugs.

The DNA-based single molecule super-resolution imaging approach, DNA-PAINT, can achieve nanometer resolution of single targets. However, the approach can suffer from significant non-specific background signals originating from non-specifically bound DNA-conjugated DNA-PAINT secondary antibodies as shown here. Using dye-modified oligonucleotides the location of DNA-PAINT secondary antibody probes can easily be observed with widefield imaging prior to beginning a super-resolution measurement.

Purine-pyrimidine repeats (PPRs) can form left-handed Z-form DNA and induce DNA double-strand breaks (DSBs), posing a risk for genomic rearrangements and cancer. The zinc finger (ZF) and BTB domain-containing protein 43 (ZBTB43) is a transcription factor containing two Cys2-His2 (C2H2) and one C3H1 zinc fingers and plays a crucial role in maintaining genomic and epigenomic integrity by converting mutagenic Z-form PPRs to the B-form in prospermatogonia. Despite its importance, the molecular mechanism underlying the recognition of PPRs by ZBTB43 remains elusive.
