The binding component (C2II) of the binary Clostridium botulinum C2 toxin mediates transport of the actin ADP-ribosylating enzyme component (C2I) into the cytosol of target cells. C2II (80 kDa) is activated by trypsin cleavage, and proteolytically activated C2II (60 kDa) oligomerizes to heptamers in solution. Activated C2II forms channels in lipid bilayer membranes which are highly cation selective and voltage-gated. A role for this channel in C2I translocation across the cell membrane into the cytosol is discussed. Amino acid residues 303-331 of C2II contain a conserved pattern of alternating hydrophobic and hydrophilic residues, which likely facilitates membrane insertion and channel formation by creating two antiparallel beta-strands. Some of the residues are in strategic positions within the putative C2II channel, in particular, glutamate 307 (E307) localized in its center and glycine 316 (G316) localized on the trans side of the membrane. Here, single-lysine substitutions of these amino acids and the double mutant E307K/G316K of C2II were analyzed in vivo and in artificial lipid bilayer experiments. The pH dependence of C2I transport across cellular membranes was altered, and a pH of

Developing the mechanism for MOF formation is crucial for the rapid development of new materials. This work demonstrates that Deuterium-NMR spectroscopy is the optimal inter-laboratory methodology for understanding the in-situ kinetics of metal-organic framework (MOF) formation. This method is facile, affordable, and allows for the isolation and monitoring of individual reagents by using one deuterated component while the remaining components are protonated.

Purpose Of Review: The role of the lymphatic system in clearing extravasated fluids, lipid transport, and immune surveillance is well established, and lymphatic vasculature can provide a vital role in facilitating crosstalk among various organ systems. Lymphatic vessels rely on intrinsic and local factors to absorb and propel lymph from the interstitium back to the systemic circulation. The biological implications of local influences on lymphatic vessels are underscored by the exquisite sensitivity of these vessels to environmental stimuli.

Rate coefficients for ion-polar-molecule reactions between acetonitrile molecules (CHCN) and nitrogen molecular ions (N), which are of importance to the upper atmospheric chemistry of Saturn's moon Titan, were measured for the first time at low translational temperatures. In the experiments, the reaction between sympathetically cooled N ions embedded in laser-cooled Ca Coulomb crystals and velocity-selected acetonitrile molecules generated using a wavy Stark velocity filter was studied to determine the reaction rate coefficients. Capture rate coefficients calculated by the Su-Chesnavich approach and by the perturbed rotational state theory considering the rotational state distribution of CHCN were compared to the experimental rate coefficients.

Alkaline water (HO) electrolysis is currently a commercialized green hydrogen (H) production technology, yet the unsatisfactory hydrogen evolution reaction (HER) performance severely limits its energy conversion efficiency and cost reduction. Herein, PtRuFeCoNi high entropy alloys (HEAs) is synthesized and subsequently exploited electrochemically induced structural oxidation processes to construct self-reconfigurable HEAs, as an efficient alkaline HER catalyst. The optimized self-reconstructed PtRuFeCoNi HEAs with the HEAs and cobalt rutheniate interface (HEAs-CoRuO) exhibits excellent alkaline HER performance, requiring just 11.

Exploiting supramolecular secondary building units (SSBUs) for developing porous crystalline materials represents an exciting breakthrough that extends the boundaries of reticular chemistry. However, shaping polynuclear clusters sustained by non-covalent interactions for the assembly of hydrogen-bonded frameworks remains a critical challenge. This study presents a novel strategy to stabilize SSBUs by tuning the π-stacking geometry of conjugated building blocks, facilitating the creation of hydrogen-bonded frameworks with tailored architectures for demanding gas separation.
