A new method for functionalization of oligonucleotides by addition of aminoalkyl derivatives to the intermolecular phosphorus atom of the oligonucleotide N3'-P5' phosphoramidate bond in the presence of triphenylphosphine, 4-dimethylaminopyridine, and 2,2'-dipyridyl disulfide was suggested. The reaction proceeded with both low-molecular alkylamines (1,6-diaminohexane or N,N-dimethyl-1,3-diaminopropane) and a ligand in minor groove containing a aminoalkyl group.

We show that a small biotin-binding RNA aptamer that folds into a pseudoknot structure acts as a substrate for bacterial RNase P RNA (RPR) with and without the RNase P C5 protein. Cleavage in the single-stranded region in loop 1 was shown to depend on the presence of a RCCA-motif at the 3' end of the substrate. The nucleobase and the 2'hydroxyl at the position immediately 5' of the cleavage site contribute to both cleavage efficiency and site selection, where C at this position induces significant cleavage at an alternative site, one base upstream of the main cleavage site.

Objective: To establish a rapid detection method for canine using recombinase-aided amplification (RAA) technology.
Methods: The outer membrane protein 25 gene fragment (Omp25) of canis was targeted. Primers and fluorescent probes were designed and synthesized, and recombinant plasmids were constructed as standards.

Synthetic ssDNA oligonucleotides hold great potential for various applications, including DNA aptamers, DNA digital data storage, DNA origami, and synthetic genomes. In these contexts, precise control over the synthesis of the ssDNA strands is essential for generating combinatorial sequences with user-defined parameters. Desired features for creating synthetic DNA oligonucleotides include easy manipulation of DNA strands, effective detection of unique DNA sequences, and a straightforward mechanism for strand elongation and termination.

Background: Spinal muscular atrophy (SMA) is a motor neuron disease caused by mutations in the SMN1 gene. Nusinersen, an antisense oligonucleotide, has been shown to improve motor function in SMA patients. However, concerns regarding its renal safety remain as previous studies have linked similar treatments to renal toxicity.

Point-of-care testing (POCT) of trace amount of biomarkers in biofluids is critical towards health monitoring and early diagnosis. In particular, to facilitate non-invasive saliva testing, the development of low-cost, lightweight and disposable biosensors is in urgent need, while the ultrahigh sensitivity beyond conventional clinical tests remains a great challenge. Herein, we demonstrate a simple and fully printable all-polymer organic electrochemical transistor (OECT) biosensor to detect femtomolar (fM)-level biomolecules in saliva within a few minutes by employing highly conducting lithium bis(trifluoromethanesulfonyl)imide (LiTFSI)-doped poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate) (PEDOT:PSS) serving as both the channel and gate.
