The development of a series of novel indole-based inhibitors of 5'-inosine monophosphate dehydrogenase (IMPDH) is described. Various hydrogen bond acceptors at C-3 of the indole were explored. The synthesis and the structure-activity relationships (SARs) derived from in vitro studies are outlined.

The current research presents novel library of indole derived fused triazole-thiadiazole derivatives (1-17) for treatment of diabetes mellitus. These compounds were synthesized by treating 2-(1H-indol-3-yl)acetic acid with hydrazinecarbothiohydrazide followed by treating the resultant compound with substituted benzoic acid. Structural validation was achieved spectroscopically (HNMR, CNMR and HREI-MS).

Bro̷nsted Acid-Catalyzed Regioselective [5 + 1] Annulation for the Synthesis of Indole-Fused Dihydrochromanes.

A Bro̷nsted acid-catalyzed [5 + 1] annulation of 2-(1-indol-2-yl)phenols, a class of indole-based 1,5-CO-synthons, with aldehydes has been established. By this approach, various indole-fused dihydrochromanes were synthesized in moderate to good yields (up to 99%) with excellent regioselectivities. Control experiments not only showed the H-bonding interaction between the NH group of the substrates─and the acid catalyst is vital to realize this transformation─but also indicated that an indolylmethanol-type intermediate was possibly formed via the first step of nucleophilic addition of indole C3-position to aldehydes during the reaction process.

Cyclooxygenase-2 (COX-2), a key enzyme in the cyclooxygenase family, is pivotal in producing pro-inflammatory prostaglandins, driving chronic inflammation and related disorders. Targeting COX-2 with selective inhibitors can mitigate these conditions while avoiding the gastrointestinal and hepatotoxic/nephrotoxic side effects of traditional NSAIDs. However, the selectivity towards COX-2 inhibition has been associated with cardiovascular risks, necessitating the discovery of novel molecular scaffolds avoiding CVS side effects.

Mechanochemical Synthesis of Carbazole Isomer Phosphors.

The 1H-benzo[f]indole (Bd[f]), a carbazole (Cz) isomer is first reported as the source of Cz-based phosphors in 2020. In this work, the novel carbazole isomers, 1H-benzo[g]indole (Bd[g]) based derivatives, are synthesized by a one-step solvent-free mechanical ball milling reaction, establishing a facile, efficient, and environmentally friendly method for the synthesis of new Cz isomer phosphorescent derivatives with high yields compared to previously reported multi-step solvent-based thermochemical synthesis routes of Bd[f] derivatives with low yields. Six Bd[g] derivatives with different substituents, namely OCH-Bd, In-Bd, Bn-Bd, F-Bd, Cl-Bd, and Br-Bd, are synthesized, which exhibit distinctly different single-crystal structures and phosphorescent properties.
