A new reactivity-based one-pot synthesis of N-acetyllactosamine oligomers.

Poly-N-acetyllactosamine oligomer is a type-2 glycan core from which a number of important bioactive glycoconjugates are assembled in vivo. Development of an effective synthesis of N-acetyllactosamine oligomers will therefore provide a new chemoenzymatic entry to this class of complex saccharides. This paper describes the design and synthesis of thioglycoside building blocks, determination of their relative reactivity values, and demonstration of their use in the programmable one-pot synthesis of various N-acetyllactosamine oligomers. Through a combination of segment condensation, the strategy allows for the preparation of larger oligosaccharides with minimal protecting group manipulation, as illustrated in the synthesis of an octasaccharide in a very short period of time.

Prolactin induced-protein (PIP) has been found to be rich in immunomodulatory epitopes, including -acetyllactosamine (LacNAc) and ,-diacetyllactosamine (LacdiNAc) residues, which may constitute ligands for galecin-3 (Gal-3). In the current study, we aimed to investigate the reactivity of galactose- and -acetylgalactosamine-specific lectins with human seminal plasma PIP. Subsequently, we examined the direct interaction between seminal plasma PIP and galectin-3, and next analyzed whether there are any differences in the interaction associated with impaired semen parameters.

Diet is one of the main factors shaping the human microbiome, yet our understanding of how specific dietary components influence microbial consortia assembly and subsequent stability in response to press disturbances - such as increasing resource availability (feeding rate) - is still incomplete. This study explores the reproducible re-assembly, metabolic interplay, and compositional stability within microbial consortia derived from pooled stool samples of three healthy infants. The mechanisms responsible for the formation and degradation of intracellular FNGs are well characterized in mammalian cells.
