Molecular analysis of prostate cancer specimens is complicated by tumor heterogeneity and admixture of malignant cells with benign epithelium, stroma and inflammatory cells. Laser Capture Microdissection allows procurement of selected groups of cells from specific areas of tissue sections. We have used Laser Capture Microdissection to obtain pure samples of malignant prostate epithelial cells from frozen radical prostatectomy specimens. Methods for DNA extraction, androgen receptor amplification and exon sequencing have been optimized. These methods should be adaptable for molecular analysis of DNA, RNA and protein from other complex tissues.

Anticounterfeiting technologies meet challenges in the Internet of Things era due to the rapidly growing volume of objects, their frequent connection with humans, and the accelerated advance of counterfeiting/cracking techniques. Here, we, inspired by biological fingerprints, present a simple anticounterfeiting system based on perovskite quantum dot (PQD) fingerprint physical unclonable function (FPUF) by cooperatively utilizing the spontaneous-phase separation of polymers and selective in situ synthesis PQDs as an entropy source. The FPUFs offer red, green, and blue full-color fingerprint identifiers and random three-dimensional (3D) morphology, which extends binary to multivalued encoding by tuning the perovskite and polymer components, enabling a high encoding capacity (about 10, far surpassing that of biometric fingerprints).

Computational microscopy with coherent diffractive imaging and ptychography.

Nature

Department of Physics and Astronomy, University of California, Los Angeles, Los Angeles, CA, USA.

Microscopy and crystallography are two essential experimental methodologies for advancing modern science. They complement one another, with microscopy typically relying on lenses to image the local structures of samples, and crystallography using diffraction to determine the global atomic structure of crystals. Over the past two decades, computational microscopy, encompassing coherent diffractive imaging (CDI) and ptychography, has advanced rapidly, unifying microscopy and crystallography to overcome their limitations.

Comprehending the biosensing mechanism of the biosensor interface is crucial for sensor development, yet accurately reflecting interfacial interactions within actual detection environments remains an unsolved challenge. An operando photoelectrochemical surface-enhanced Raman spectroscopy (PEC-SERS) biosensing platform was developed, capable of simultaneously capturing photocurrent and SERS signals, allowing operando characterization of the interfacial biosensing behavior. Porphyrin-based MOFs (Zr-MOF) served as bifunctional nanotags, providing a photocurrent and stable Raman signal output under 532 nm laser irradiation.

Endometrial decidualization resistance (DR) is implicated in various gynecological and obstetric conditions. Here, using a multi-omic strategy, we unraveled the cellular and molecular characteristics of DR in patients who have suffered severe preeclampsia (sPE). Morphological analysis unveiled significant glandular anatomical abnormalities, confirmed histologically and quantified by the digitization of hematoxylin and eosin-stained tissue sections.

In orchard environments, negative obstacles such as ditches and potholes pose significant safety risks to robots working within them. This paper proposes a negative obstacle detection method based on LiDAR tilt mounting. With the LiDAR tilted at 40°, the blind spot is reduced from 3 m to 0.
