Antiviral properties of prodelphinidin B-2 3'-O-gallate from green tea leaf.

Prodelphinidin B-2 3-O-gallate, a proanthocyanidin gallate isolated from green tea leaf, was investigated for its anti-herpes simplex virus type 2 properties in vitro. Prodelphinidin B-2 3'-O-gallate exhibited antiviral activity with IC50 of 5.0 +/-1.0 microM and 1.6 +/-0.3 pM for XTT and plaque reduction (PRA) assays, respectively. Cytotoxicity assay had shown that prodelphinidin B-2 3'-O-gallate possessed cytotoxic effect toward Vero cell at concentration higher than its IC50. The 50% cytotoxic concentration for cell growth (CC50) was 33.3 +/- 3.7 microM. Thus, the selectivity index (SI) (ratio of IC50 to CC50) for XTT assay and PRA was 6.7 and 20.8, respectively. Prodelphinidin B-2 3'-O-gallate significantly reduced viral infectivity at concentrations 10 microM or more. Result of time-of-addition studies suggested that prodelphinidin B-2 3'-O-gallate affected the late stage of HSV-2 infection. In addition, it was also shown to inhibit the virus from attaching and penetrating into the cell. Thus, prodelphinidin B-2 3'-O-gallate was concluded to possess antiviral activity with mechanism of inhibiting viral attachment and penetration, and disturbing the late stage of viral infection.

Two new A-type dimeric prodelphinidins, EGC-(2β→O7, 4β→8)-C and EGC-(2β→O5, 4β→6)-C, were isolated from the skins of roasted hazelnut ( Corylus avellana L.) by low-speed rotary countercurrent chromatography (LSRCCC) and final purification by preparative HPLC. Their structures were determined by a combination of mass spectrometry (HPLC-ESI-MS(n) and HR-ESI-MS) and NMR spectroscopy that included the application of 2D methods ((1)H-(1)H COSY, HSQC, HMBC, and NOESY).

The pattern of the monomeric and oligomeric flavan-3-ols for 10 barley varieties and the corresponding malts were identified and quantified using high-performance liquid chromatography-ultraviolet detection-electrospray ion trap mass spectrometry. The Folin-Ciocalteau and the vanillin spectrophotometric assays were used for the assessment of the total polyphenol and total flavan-3-ol content, respectively, and the antioxidant activity was determined as the 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and the ferricyanide reducing power. Catechin and prodelphinidin B3 were respectively the major monomeric and dimeric flavan-3-ols.

Much is known about the bioactive properties of green tea flavan-3-ol. However, very little work has been done to determine the properties of proanthocyanidins, another kind of polyphenols in green tea. In this study, we have investigated the anti-inflammatory effect of tea prodelphinidin B-4 3'-O-gallate (PDG) by demonstrating the inhibitory effects on cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-activated murine macrophage RAW264 cells.

Prodelphinidin B-2 3'-O-gallate, a proanthocyanidin gallate isolated from green tea leaf, was investigated for its anti-proliferative activity in human non-small cell lung cancer A549 cells. The results showed that prodelphinidin B-2 3'-O-gallate inhibited the proliferation of A549 cells with no detectable toxic effects on normal WI-38 cells as measured by the XTT assay. Flow cytometric analysis showed that prodelphinidin B-2 3'-O-gallate blocked cell cycle progression in the G0/G1 phase.
