In contrast to the conventional dogma that carbohydrates are poorly immunogenic T-cell-independent antigens, zwitterionic polysaccharides (ZPSs) can significantly stimulate T-cell proliferation and regulate abscess formation in bacterial infection. Despite their similar biological activities, ZPSs from various bacteria are greatly different in primary chemical compositions and building block linkages. To identify the common structural features that govern the peculiar immunologic activity of ZPSs, we have been determining three-dimensional structures of compositionally different ZPSs by NMR spectroscopy and molecular mechanics and dynamics calculations. We report here the conformation of type 1 capsular polysaccharide from the human pathogen Streptococcus pneumoniae (Sp1) to be a right-handed helix with repeated zwitterionically charged grooves. We also report the striking similarity between the structures of Sp1 and our previously determined PS A2 from Bacteroides fragilis. These results support our hypothesis that T-cell-activating ZPSs assume similar conformational and charge patterns that are recognized by specific receptors and that account for their common property as T-cell activators.

Hydrogel indicators promise to monitor food spoilage, but their poor mechanics can cause defects in transport. Herein, a novel zwitterionic double network (DN) hydrogel was developed by polymerizing arylamide and sulfobetaine methacrylate in an alginate-Ca system. This hydrogel exhibited enhanced mechanical properties, including a maximum 2087 % breaking elongation and 135 ± 12 kJ/m toughness, significantly outperforming the current zwitterionic DN hydrogels, which typically exhibit less than 1800 % breaking elongation, capable of supporting 150 g-136 times its own weight.

Background/objectives: The MHCII-dependent, CD4+ T-cell zwitterionic polysaccharide PS A1 has been investigated as a promising carrier for vaccine development because it can induce an MHCII-dependent CD4+ response towards a variety of tumor-associated carbohydrate antigens (TACAs). However, PS A1 cannot elicit cytotoxic T lymphocytes through MHCI, which may or may not hamper its potential clinical use in cancer, infectious and viral vaccine development. This paper addresses PS A1 MHCI independence through the introduction of an MHCI epitope, the poliovirus (PV) peptide, to establish an MHCI- and MHCII-dependent vaccine.

The simultaneous separation of the enantiomers of six anticoagulant rodenticides, derived from 4-hydroxycoumarin, has been studied in this work. Ten different stationary phases (zwitterionic, Pirkle-type, polysaccharides and macrocyclic antibiotics derivatives) were evaluated by using supercritical fluid chromatography coupled to two different detectors (circular dichroism and mass spectrometry-single quadrupole). The effect of the type of organic modifier and temperature on the chiral separation was investigated, and the best results were obtained with the column Regis S,S-Whelk-O1 at 25 °C when using a gradient elution program with methanol as organic modifier.
