Dextro-amphetamine increases phosphoinositol cycle activity in volunteers: an MRS study.

Published: December 2002

Aims: To help determine the effects of dextro-amphetamine on the phosphoinositol cycle (PI-cycle) in humans, (1)H and (31)P magnetic resonance spectroscopy (MRS) was utilized in 17 healthy volunteers. This was an open-label study carried out before and after administration of 20 mg oral dextro-amphetamine. Subjects also rated the subjective effects of dextro-amphetamine administration using visual analog scales (VAS).

Major Findings: Following dextro-amphetamine administration there was a significant increase in the concentrations of both myo-inositol and phosphomonoesters.

Conclusions: These findings are in keeping with suggestions that dextro-amphetamine administration increases the activity of the phosphoinositol cycle, probably via an indirect release of dopamine and noradrenaline. These results are the first time that this has been confirmed in humans.

Objective: In this study, a high-throughput sequencing technology was used to screen the differentially expressed miRNA in the patients with "fast" and "slow" progression of chronic obstructive pulmonary disease (COPD). Moreover, the possible mechanism affecting the progression of COPD was preliminarily analyzed based on the target genes of candidate miRNAs.

Methods: The "fast" progressive COPD group included 6 cases, "slow" and Normal progressive COPD groups included 5 cases each, and COPD group included 3 cases.

Myo-inositol is a natural polyol, the most abundant among the nine possible structural isomers available in living organisms. Inositol confers some distinctive traits that allow for a striking distinction between prokaryotes and eukaryotes, the basic clusters into which organisms are partitioned. Inositol cooperates in numerous biological functions where the polyol participates or by furnishing the fundamental backbone of several related derived metabolites, mostly obtained through the sequential addition of phosphate groups (inositol phosphates, phosphoinositides, and pyrophosphates).

Reduced sulfatide content in deferoxamine-induced senescent HepG2 cells.

Iron chelators, such as deferoxamine, exert an anticancer effect by altering the activity of biomolecules critical for regulation of the cell cycle, cell metabolism, and apoptotic processes. Thus, iron chelators are sometimes used in combination with radio- and/or chemotherapy in the treatment of cancer. The possibility that deferoxamine could induce a program of senescence similar to radio- and/or chemotherapy, fostering adaptation in the treatment of cancer cells, is not fully understood.
