A data-collection method for macromolecular crystals using convergent sources is described here. Because of the unique characteristics of the diffraction patterns, a software package CBMPRO has been developed specifically for processing data images collected with the convergent beam method (CBM). The resulting data sets from crystals with two different sets of unit-cell parameters are presented and compared. There is good agreement between data sets from the same type of crystals under slightly different experimental conditions and data sets collected and processed with CBM also agree well with those from conventional oscillation methods, marking an important step to establishing CBM as a viable alternate data-collection method for macromolecular crystals.

Background: Hallmark pathologies of Alzheimer's Disease (AD) include the accumulation of both extracellular amyloid and intracellular tau proteins. Crystal structure determination is a crucial aspect of almost every branch of the chemical sciences, bringing us closer to understanding crystallization, polymorphism, phase transitions, and the relationship between a structure and its physicochemical and functional properties. Current treatments for fundus disorders, such as intravitreal injections, pose risks, including infection and retinal detachment, and are limited in their ability to deliver macromolecular drugs across the blood‒retinal barrier. Although non-invasive methods are safer, their delivery efficiency remains suboptimal (<5%). We have developed a wearable electrodriven switch (WES) that improves the non-invasive delivery of macromolecules to the fundus.

In this work a novel method for synthesis of 1,8-dihydroxynaphthalene melanin was presented, as well as the physicochemical properties, molecular structure, and characteristics of the pigment. The proposed synthesis protocol is simple and cost-effective with no enzymes or catalysts needed. The final product is not adsorbed on any surface, since the pigment is the result of autooxidation of 1,8-dihydroxynaphthalene.
