Purpose: The purpose of this study was to characterize the transport of uracil, a pyrimidine nucleobase, in Caco-2 and LLC-PK, cells.

Methods: Caco-2 and LLC-PK1 cells were grown to confluency on a permeable polycarbonate membrane insert to permit transport and uptake experiments after the loading of uracil on either the apical or basolateral side.

Results: The vectorial transport of uracil in both directions was saturable with comparable Km and Vmax in Caco-2 cell monolayers, probably because of a Na+-independent transport system located on the basolateral membrane. In LLC-PK1 cell monolayers, two distinct transport systems, namely a Na+-dependent and a Na+-independent, were functional in the apical to basolateral (A-B) transport of uracil. The first system was saturable with a Km value of 3.67 +/- 0.40 microM, a Vmax of 11.31 +/- 0.91 pmol/cm2/min, and a Na+:uracil coupling stoichiometry of 1.28 +/- 0.20. The second system was Na+ independent and satuable with a low affinity (Km, 50.37 +/- 9.61 microM) and Vmax (16.01 +/- 4.48 pmol/cm2/min). The two transport systems appeared to be located on the apical membrane.

Conclusion: The mechanism of uracil transport differs depending on cell lines; a Na+-independent system on the basolateral membrane in Caco-2 cells and both Na+-dependent and Na+-independent systems on the apical membrane in LLC-PK1 cells seem to be responsible for the difference.

Chemotherapy resistance is a great challenge in the treatment of gastric cancer (GC), so it is urgent to explore the prognostic markers of chemoresistance. PUF60 (Poly (U)-binding splicing factor 60) is a nucleic acid-binding protein that has been shown to regulate transcription and link to tumorigenesis in various cancers. However, its biological role and function in chemotherapy resistance of GC is unclear.

Gastric cancer (GC) ranks as the fifth leading cause of cancer mortality, with cancer stem cells (CSCs) playing a critical role in tumor progression and resistance to chemotherapy. Conventional chemotherapy often fails to effectively target these stem cells. BATF2, a tumor suppressor, is known for its role in gastric cancer, but its influence on cancer stem cell-like properties and chemotherapy response remains unclear.

Although combining 5-fluorouracil (5-FU) and Interferon-beta (IFNb) improves response rates in Hepatocellular Carcinoma (HCC), the outcomes remain suboptimal. This study investigates the feasibility of using highly transfected Mesenchymal Stem Cells (MSCs) to deliver a chemotherapeutic (5-FU) and an immunomodulator (IFNb) for localized HCC treatment. Considering the crucial role of cold-chain transportation in off-the-shelf allogeneic therapy, the study also assesses the quality and efficacy of frozen-thawed engineered MSCs, simulating a multisite study process.

Effective drug release is of utmost importance in the medical field for treating various diseases, particularly cancer. Nanocomposite hydrogels remain the best materials for enhancing the bioavailability and therapeutic levels of drugs as they enable sustained, targeted, or controlled drug release. In this work, a nanocomposite hydrogel containing locust bean gum (LBG), poly(4-acryloylmorpholine) (PAcM), and silver nanoparticles (SN) has been made using an eco-friendly microwave (MW)-assisted method and characterized by various advanced techniques.
