Reaction between ethyl 3-N,N-(dimethylamino)-2-isocyanoacrylate (1) and a primary amine (2) regioselectively affords 1-alkyl-4-imidazolecarboxylates (3) in good yields (52-89%). The method is applicable to unhindered and hindered amine substrates, as well as those containing reactive functionality such as alcohols and secondary and tertiary amines. [reaction: see text]

Despite tremendous progress of dienamine catalysis along with its application in enantioselective synthesis over nearly two decades, certain limitations, especially with respect to the regioselectivity in the dienamine generation step, continue to persist. To overcome these shortcomings of classical dienamine catalysis, we now introduce the concept of alkoxy-directed dienamine catalysis and apply it to the enantioselective arene construction by desymmetrizing -enediones through [4 + 2]-cycloaddition. Catalyzed by a diphenylprolinol silyl ether, this reaction utilizes γ-alkoxy α,β-unsaturated aldehydes as the substrate and proceeds in a highly regioselective fashion through the intermediacy of δ-alkoxy dienamine.

In the chemical synthesis or modification of saccharides, regioselective protection of the many similar OH groups in saccharides is necessary but remains a major challenge. In particular, the regio- and stereoselective conversion of C(1,2)-OH has great synthetic potential in carbohydrate synthesis but has largely remained untapped. Here, an in situ proton-producing system mediated by boronic acid was found and employed for site-selective ketalization of various unprotected saccharides.

The electron donors (ED) in Ziegler-Natta (Z-N) catalysis are classified as internal electron donors (IED) and external electron donors (EED), and both IED and EED are indispensable components for enhancing the catalytic reactivity and regulating the stereoregularity of polyolefinic materials in a typical industrial Z-N catalytic process. With the intensive research on ED, the Z-N catalyst performances have experienced successive progress in the last few decades. Polypropylenes (PP) as a commodity polyolefin material, especially the isotactic PP (iPP), are produced in multi-billion pounds per annum by utilization of the various IED- and EED-assisted Z-N catalysts systems.

The bisbenzylisoquinoline alkaloids (bisBIAs) have attracted tremendous attention from the synthetic community due to their diverse and intriguing biological activities. Herein, we report the convergent and modular chemoenzymatic syntheses of eight bisBIAs bearing various substitutes and linkages in 5-7 steps. The gram-scale synthesis of various well-designed enantiopure benzylisoquinoline monomers was accomplished through an enzymatic stereoselective Pictet-Spengler reaction, followed by regioselective enzymatic methylation or chemical functionalization in a sequential one-pot process.
