Functional analysis of the Escherichia coli zinc transporter ZitB.

The membrane transporter ZitB responsible for Zn(II) efflux in Escherichia coli was studied by site-directed mutagenesis to elucidate the function of individual amino acid residues. Substitutions of several charged or polar residues, H53, H159, D163 and D186, located in predicted transmembrane domains resulted in loss of ZitB function. In contrast, neither the amino-terminal nor the carboxy-terminal regions, both histidine-rich, were required for function.

Indigo is widely used in dyes, medicines and semiconductors materials due to its excellent dyeing efficiency, antibacterial, antiviral, anticancer, anti-corrosion, and thermostability properties. Here, a biosynthetic pathway for indigo was designed, integrating two enzymes (EcTnaA, MaFMO) into a higher L-tryptophan-producing the strain Escherichia coli TRP. However, the lower catalytic activity of MaFMO was a bottleneck for increasing indigo titers.

Background: Lower urinary tract disease is a common clinical condition in dogs, usually presenting with dysuria, pollakiuria and haematuria. Diabetes mellitus is a predisposing factor for urinary tract infection in both humans and dogs and does not necessarily present with clinical signs.
