Purpose: Due to the slowly progressing nature of age-related macular degeneration (AMD) and critical differences in ocular anatomy between humans and animals, it has been difficult to model disease progression, hampering the development of novel therapeutics aimed at impacting drusen biogenesis. To determine whether "drusen-in-a-dish" model systems are of utility in screening potential therapeutics aimed at early-intermediate dry AMD, we developed a detailed characterization of the protein, glycoprotein, and lipid composition of sub-retinal pigment epithelium (RPE) deposits grown by monolayers of ex vivo porcine RPE with human drusen in AMD globes.
Methods: Immunohistochemistry and imaging mass spectrometry (IMS) were performed on 20-week aged monolayers of porcine RPE and human donor globes recovered from an 81-year-old non-transplant donor with confirmed diagnosis of bilateral dry AMD.

Background: Circadian rhythm disorders and NF-κB are closely linked and can exacerbate periodontitis. However, the mechanisms via which circadian rhythm-related genes influence periodontitis are not yet fully understood.
Objective: We investigated the effect of brain and muscle Arnt-like protein-1 (BMAL1) on the NF-κB pathway and downstream inflammatory factors on periodontitis.

The boxwood leafminer (Diptera, Cecidomyiidae) has historically been considered a leafminer, but some researchers suggested it induced galls on species leaves. The larvae of create small blister-like galls on leaves, causing tissue hypertrophy and hyperplasia. Histological examination reveals that larvae cause the formation of small blister galls, which involve tissue reorganization in the mesophyll.

In recent years, mass spectrometry-based imaging techniques have improved at unprecedented speeds, particularly in spatial resolution, and matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI) experiments can now routinely image molecular profiles of single cells in an untargeted fashion. With the introduction of MALDI-immunohistochemistry (IHC), multiplexed visualization of targeted proteins in their native tissue location has become accessible and joins the suite of multimodal imaging techniques that help unravel molecular complexities. However, MALDI-IHC has not been validated for use with cell cultures at single-cell level.

Adipose tissue is a highly dynamic endocrine organ that serves as the body's primary energy reservoir through the storage and mobilization of lipids. Adipocyte cellular size has been recognized as an indicator of cellular status; hypertrophic adipocytes are more prone to insulin resistance and the secretion of pro-inflammatory cytokines. Thus, the size and number of lipids is important to consider both in the clinic with a biopsy and when developing disease models and regenerative tissue constructs.
