alpha-Lipoic acid (LA), an antioxidant with broad neuroprotective capacity, is thought to act by scavenging reactive oxygen species and stimulation of glutathione synthesis. LA shows structural resemblance to dithiolethiones, like anethole dithiolethione (ADT). ADT protects against oxidative damage, primarily by induction of phase II detoxication enzymes, in particular NAD(P)H:quinone oxidoreductase (NQO1) and glutathione-S-transferase (GST). Therefore, we investigated whether LA, like ADT, is capable also of inducing these protective enzymes. Our data show that LA, like ADT, induces a highly significant, time- and concentration dependent, increase in the activity of NQO1 and GST in C6 astroglial cells. The LA or ADT mediated induction of NQO1 was further confirmed by quantitative PCR and western blot analysis. This work for the first time unequivocally demonstrates LA mediated upregulation of phase II detoxication enzymes, which may highly contribute to the compounds' neuroprotective potential. Moreover, the data support the notion of a common mechanism of action of LA and ADT.

Hardy plants play a crucial role in restoring high-altitude tailings ponds, but the accumulation of potentially toxic elements (PTEs) and detoxification mechanisms in alpine plants are understudied. This study first investigated the cadmium (Cd) accumulation capacity and detoxification mechanisms by comparative transcriptomics with different Cd stress (0, 5, 10, 20 and 40 mg L Cd) of Koenigia tortuosa from a lead-zinc mine (4950 m above sea level) in Qinghai-Tibet Plateau. The findings revealed that, despite elevated Cd concentrations suppressed the growth of Koenigia tortuosa, the plant retained a notable ability to accumulate Cd.

Ethnopharmacological Relevance: Emilia sonchifolia is a very widely used traditional Chinese medicine, with the efficacy of heat-clearing, detoxicating, dissipating blood stasis, reducing swelling and relieving pain. As a widely used traditional miao herb, Emilia sonchifolia is often used to treat upper respiratory tract infections, oral ulcer, pneumonia, mastitis, enteritis, bacillum, urinary tract infection, sores, eczema, falls and injuries, etc. In fact, many cases of liver injury caused by Emilia sonchifolia have been reported clinically.

Glutathione S-transferases (GSTs) are evolutionarily conserved enzymes crucial for cell detoxication. They are viewed as having evolved in cyanobacteria, the ancient photosynthetic prokaryotes that colonize our planet and play a crucial role for its biosphere. Xi-class GSTs, characterized by their specific glutathionyl-hydroquinone reductase activity, have been observed in prokaryotes, fungi and plants, but have not yet been studied in cyanobacteria.

Nanozymes, a category of nanomaterials with exceptional enzyme-like activity, exhibit the significant promise to overcome the inherent limitations of natural enzymes. Inspired by the active site structure of natural laccase, a biomimetic MA-Cu nanozyme with three-dimensional network structure was constructed in water system through one-step complexation based on the specific coordination between nitrogen-rich triazine heterocyclic melamine and Cu, in a facile, green and economical manner. Compared to natural laccase, MA-Cu possesses superior multi-enzyme mimicking activity, stability and cost-effectiveness.

Glutathione transferases are detoxication enzymes with broad catalytic diversity, and small alterations to the protein's primary structure can have considerable effects on the enzyme's substrate selectivity profile. We demonstrate that two point mutations in glutathione transferase P1-1 suffice to generate 20-fold enhanced non-selenium-dependent peroxidase activity indicating a facile evolutionary trajectory. Designed mutant libraries of the enzyme were screened for catalytic activities with alternative substrates representing four divergent chemistries.
