Three-dimensional structure of human tubulin chaperone cofactor A.

alpha and beta-Tubulin fold in a series of chaperone-assisted steps. At least five protein cofactors are involved in the post-chaperonin tubulin folding pathway and required to maintain the supply of tubulin; some of them also participate in microtubule dynamics. The first tubulin chaperone identified in the tubulin folding pathway was cofactor A (CoA). Here we describe the three-dimensional structure of human CoA at 1.7 A resolution, determined by multiwavelength anomalous diffraction (MAD). The structure is a monomer with a rod-like shape and consists of a three-alpha-helix bundle, or coiled coil, with the second helix kinked by a proline break, offering a convex surface at one face of the protein. The helices are connected by short turns, one of them, between alpha2 and alpha3, including a 3(10)-helix. Peptide mapping analysis and competition experiments with peptides show that CoA interacts with beta-tubulin via the three alpha-helical regions but not with the rod-end loops. The main interaction occurs with the middle kinked alpha2 helix, at the convex face of the rod. Strong 3D structural homology is found with the Hsp70 chaperone cofactor BAG domain, suggesting that these proteins define a family of cofactors of simple compact architecture. Further structural homology is found with alpha-spectrin/alpha-actinin repeats, all are rods of identical length of ten helical turns. We propose to call these three-helix bundles alpha ten modules.

This work developed a novel oxidized hierarchical porous carbon (OHPC) with vesicule-like ultrathin graphitic walls via a method of air oxidation and used as an efficient adsorbent for Congo red (CR) and Malachite green (MG) removal. Results show that the OHPC2 oxidized at 400 °C possesses three-dimensional hierarchical pores with vesicule-like ultrathin graphitic walls. The prepared OHPC2 not only has a large specific surface area of 1020 m g with a high pore volume, but also has abundant oxygen-containing functional groups.

Background/aim: Autosomal-recessive carnitine-acylcarnitine translocase deficiency (CACTD) is a rare disorder of long-chain fatty acid oxidation caused by variants in the SLC25A20 gene. Under fasting conditions, most newborns with severe CACTD experience sudden cardiac arrest and hypotonia, often leading to premature death due to rapid disease progression. Understanding of genetic factors and pathogenic mechanisms in CACTD is essential for its diagnosis, treatment, and prevention.

A three-dimensional numerical model of the vacuum sintering furnace was established, combined with the custom program of temperature-voltage feedback regulation. Through simulationand experimental validation, the heating and holding stage as well as the thermal hysteresis phenomenon of the furnace were analyzed, a dimensionless quantity of hysteresis temperature difference was proposed and calculated, the distribution of the electric field and temperature uniformity of the furnace were discussed in detail, while the structural improvement approach was proposed based on simulation. The results show that: during the heating process, the maximum of thermal hysteresis temperature difference between the graphite cylinder and the heating tube is 0.

This study employed numerical simulation to investigate the dynamic response characteristics of open-web girders subjected to proximity blast loading and to compare these characteristics with those of solid-web girders. The research utilized the Coupled Eulerian-Lagrangian (CEL) method for simulation, effectively combining the advantages of both Eulerian and Lagrangian approaches. This method mitigated issues related to mesh distortion while accurately modeling the damage inflicted by blast loads on the structures.
