Practical advice in the development of a lyophilized protein drug product.

Antib Ther

January 2025

Formulation Development Group, Regeneron Pharmaceuticals Inc, 777 Old Saw Mill River Rd, Tarrytown, NY 10591, USA.

The development of lyophilized protein drug products is a critical and complex task in the pharmaceutical industry, requiring a comprehensive understanding of the myriad of factors affecting product quality, stability, and the efficiency and robustness of the lyophilization process. This review offers practical advice on the critical aspects of lyophilized protein drug product development. Practical considerations across both the early and late stages of development are discussed, underscoring the necessity of a strategic approach from initial development through to commercialization.

There is an increased risk of cardiac complications in COVID-19 among patients with pre-existing comorbidities due to their existing pro-inflammatory status. The manifestation of complete heart block has been evidenced as transient and in no need of intervention. This case presents a previously healthy woman who was diagnosed with SARS-CoV-2 infection and complete heart block; she underwent permanent dual-chamber pacemaker implantation after successful treatment and a negative COVID-19 test.

A 17-year-old patient presented with frequent palpitations, where the tachycardia was not sustained and could not be induced, making it impossible to pinpoint the earliest activation site using the activation map. However, by utilizing a dual-chamber electrogram-based pace mapping technique, we successfully identified the origin and achieved effective treatment.

Ppb-Level Photoacoustic Detection of Chloroform Using Four-Microphone Array.

Anal Chem

January 2025

International Joint Laboratory for Integrated Circuits Design and Application, Ministry of Education, School of Physics, Zhengzhou University, Zhengzhou 450001, China.

The photoacoustic spectroscopy (PAS) system commonly enhances the efficiency of optical-acoustic-electrical energy conversion by increasing the laser power, optimizing the resonance characteristics of the photoacoustic cell (PAC), and improving the sensitivity of acoustic sensors. However, conventional systems using a single-microphone or a dual-microphone differential setup for point sampling of the photoacoustic signal fail to account for its spatial distribution, leading to a loss of spatial gain. Drawing on microphone array theory derived from sonar technology, this study, for the first time, presents a PAS sensing system based on a four-microphone array, which is applied to detect chloroform gas.

Background: Transvenous lead extraction (TLE) has become an essential component of lead management strategies, but it carries the risk of severe complications, including damage to the tricuspid valve. Currently, there are no established predictors that can help prevent these complications.

Case Summary: An 84-year-old male with a dual-chamber pacemaker was admitted to the hospital due to a pocket fistula resulting from a local infection.
