New beta-amino alcohols possessing a vinylsilane moiety were reacted with glyoxal to produce lactones that were transformed in three steps in enantiopure pipecolic acid derivatives. The key step was a totally diastereoselective ene-iminium cyclization, whose mechanism can be viewed as a direct cyclization of the vinylsilane moiety onto the iminium ion. The reactivity of two beta-amino alcohols having an allylsilane terminator was also examined. Their difference of reactivity toward glyoxal can be ascribed to the intervention of a carbocation, which presents a behavior that differs according to the position of the trimethylsilyl group.

We synthesized a series of amphiphilic cationic poly(ethylene glycol)--poly(β-amino acid) derivatives with various lengths of PEG to investigate the effects of PEG lengths on mRNA transfection. The surface charges of the polyplexes with a DP ≥ 4 gradually decreased as DP increased. This indicated that hydrophilic PEG with a DP ≥ 4 was exposed on the surface of the polyplexes, which was further confirmed using H NMR.

This study describes a general method for the preparation of 1,4-oxazin-2-one intermediates from acetylene dicarboxylate and β-amino alcohol precursors. Oxazinones prepared in this manner were employed in a tandem cycloaddition/cycloreversion reaction sequence with a model alkyne (phenyl acetylene) to give substituted pyridine products. Fundamental reactivity and selectivity studies are complemented by the synthesis of the polycyclic ergot alkaloid natural product xylanigripone A.
