Diastereoselective methylation of dimethyl (S)-malate 7, followed by two, three-step reductive de-hydroxylation procedures afforded dimethyl (R)-2-methylsuccinate 11 in 80.2% e.e. and 84.7% e.e., respectively. The latter compound was further transformed into the natural enantiomers of the ant venom alkaloids (R)-leptothoracine 1 and (R)-3-methyl-N-(2-phenylethyl)-pyrrolidine 2.

A C-H Arylation-Based Enantioselective Synthesis of Planar Chiral Cyclophanes.

Angew Chem Int Ed Engl

University of Basel, Department of Chemistry, St. Johanns-Ring 19, 4056, Basel, SWITZERLAND.

Despite the growing importance of planar chiral macrocyclophanes owing to their unique properties in different areas of chemistry, methods that are effective in controlling their planar chirality are restricted to certain molecular scaffolds. Herein, we report the first Pd(0)-catalyzed enantioselective intermolecular C-H arylation that induces planar chirality by installing bulky aryl groups through dynamic kinetic resolution (DKR). A computer-assisted approach allowed a fine-tuning of the structure of the employed chiral bifunctional phosphine-carboxylate ligands to achieve high enantioselectivities.

Factor XIa (FXIa) is a plasma protease that plays a crucial role in the intrinsic pathway of blood coagulation, making it a promising target for antithrombotic therapy. Circular DNA aptamers, with their dramatically enhanced biological and structural stability, hold great potential as new-generation DNA-based anticoagulants. However, the functional selection and large-scale synthesis of them remains a substantial challenge.

Recent innovations in strategies to prepare metal-organic framework-based mixed matrix membranes.

Chem Commun (Camb)

January 2025

Department of Chemistry, College of Sciences, Northeastern University, Shenyang 110819, P. R. China.

Mixed matrix membranes (MMMs) composed of metal-organic frameworks (MOFs) and polymer matrixes have garnered significant attention due to their potential to overcome the permeability-selectivity trade-off inherent in polymeric membranes. Nevertheless, the application and industrial production of MOF-based MMMs have been hindered by issues such as poor interfacial compatibility and cumbersome fabrication processes. Recently, strategies have emerged as promising approaches for fabricating MOF-based MMMs, offering enhanced interfacial compatibility between MOF fillers and polymers, as well as a simplified construction process.

Chemical 1,1'-glycosylation for the synthesis of non-reducing disaccharides is complicated by the need to simultaneously control the stereochemistry at two anomeric centers. While considerable progress has been made in the synthesis of α,α-disaccharides, the assembly of 1,1'-β,β- and 1,1'-β,α-linked non-reducing sugars has received comparatively less attention. Many naturally occurring non-reducing disaccharides and their biologically active mimetics feature asymmetrically located functional groups at different positions on the two pyranose rings, highlighting the demand for reliable stereoselective methods to synthesize fully orthogonally protected 1,1'-conjugated sugars suitable for targeted functionalisation to create important biomolecules.

Superior colliculus controls the activity of the substantia nigra pars compacta and ventral tegmental area in an asymmetrical manner.

J Neurosci

Department of Neurophysiology and Chronobiology, Institute of Zoology and Biomedical Research, Jagiellonian University, 9 Gronostajowa street, 30-387 Kraków, Poland.

Dopaminergic (DA) neurons of the substantia nigra pars compacta (SNc) and ventral tegmental area (VTA) play a crucial role in controlling animals' orienting and approach behaviors toward relevant environmental stimuli. The ventral midbrain receives sensory input from the superior colliculus (SC), a tectal region processing information from contralateral receptive fields of various modalities. Given the significant influence of dopamine release imbalance in the left and right striatum on animals' movement direction, our study aimed to investigate the lateralization of the connection between the lateral SC and the midbrain DA system in male rats.
