[The ambiguity of the difference].

The article concentrates on the process of diagnosing disease, especially of common health problems. The biomedical concept dominates medicine in our days. However, the majority of the complaints presented to the general practician cannot be classified as distinct disease entities. An integral concept is far more suitable to integrate biomedical and psychosocial factors. In this way biomedical aspects acquire significance within an individual life story and social context in which gender plays a role. That is why the integral concept is more adequate to focus on sex differences in the diagnostic process. The article describes the impediments in this process, mainly due to the fact the prevailing concepts of disease ignore sex differences.

Peru is among Latin American countries with the largest Indigenous population, yet ethnical health disparities persist, particularly in the Amazon region which comprises 60% of the national territory. Healthcare models that include Indigenous medicine and traditional healers present an important avenue for addressing such inequalities, as they increase cultural adequacy of services, healthcare access, and acknowledge Indigenous Rights for their perspectives to be represented in public healthcare. Understanding the underlying epistemologies of Indigenous medicine is a prerequisite for this purpose.

Rationalizing Predictions of Isoform-Selective Phosphoinositide 3-Kinase Inhibitors Using MolAnchor Analysis.

J Chem Inf Model

January 2025

Department of Life Science Informatics and Data Science, B-IT, LIMES Program Unit Chemical Biology and Medicinal Chemistry, Rheinische Friedrich-Wilhelms-Universität, Friedrich-Hirzebruch-Allee 5/6, Bonn D-53115, Germany.

Explaining the predictions of machine learning models is of critical importance for integrating predictive modeling in drug discovery projects. We have generated a test system for predicting isoform selectivity of phosphoinositide 3-kinase (PI3K) inhibitors and systematically analyzed correct predictions of selective inhibitors using a new methodology termed MolAnchor, which is based on the "anchors" concept from explainable artificial intelligence. The approach is designed to generate chemically intuitive explanations of compound predictions.
