Feeding experiments using 14C-labelled precurosrs demonstrate the following sequence of the biological formation of penicillic acid in Penicillium cyclopium: orsellinic acid leads to 2-O-methylorsellinic acid leads to 1,4-dihydroxy-6-methoxy-2-methylbenzene leads 6-methoxy-2-methyl-benzoquinone(1,4)leads to penicillic acid.

Overexpression of a global transcriptional regulator LaeA was performed to awaken cryptic biosynthetic gene clusters (BGCs) in a marine sponge-derived fungus, Aspergillus niger L14, to obtain secondary metabolites (SMs), and resulted in the production of five previously undiscovered SMs in the wild-type strain, including three cyclic lipopeptides aspochracin (1), JBIR-15 (2) sclerotiotide C (3), kojic acid (4) and penicillic acid (5). Bioassays revealed that compound 2 displayed promising antimicrobial effects on Candida albicans with a minimum inhibitory concentration value of 32 µg/mL and 4 exhibited significant antioxidant activity with a similar half-maximal inhibitory concentration value (5 µg/mL) to that of ascorbic acid. Transcriptome analysis results indicated that the overexpression of LaeA in strain L14 remarkably enhanced the expression of genes involved in polyketide synthase-non-ribosomal peptide synthetase hybrids and amino acid metabolism, demonstrating an effective approach for the production of cyclic lipopeptides.

(1) Background: Food and feed safety legislation does not concern all the mycotoxins generated by and spp. Certain mycotoxins, including mycophenolic acid (MPA), cyclopiazonic acid (CPA), penicillic acid (PA), roquefortine C (ROQ C), and gliotoxin (GLI), are regarded as having lower toxicity levels, and hence are not included in food and feed legislation. It is obvious that xenobiotics, including mycotoxins, exert synergistic harmful health effects on human and animal when exposed through food and feed.

Alzheimer's Disease (AD) is a neurodegenerative disorder proven to be caused by the aggregation of protein tau into fibrils, resulting in neuronal death. The irreparable neuronal damage leads to irreversible symptoms with no cure; therefore, disaggregation of these tau fibrils could be targeted as a therapeutic approach to AD. Here we have developed a fungal natural product library to screen for secondary metabolites that have bioactive potential towards AD tau.

Exploration of medicinal plants for bioactive-producing endophytic fungi is a relatively unmapped source of pharmaceutically important compounds. In this study, the endophytic fungus Curvularia lunata AREF029 isolated from the medicinal plant Cymbopogon citratus (known as lemongrass) was assessed for its biological activity. The methanolic extract of AREF029 had minimum inhibition concentration (MIC) ranging from 38 to 174 µg/ml against phytopathogenic fungi Alteranria solani, Fusarium oxysporum and Rhizoctonia solani.
