Mapping mouse hemangioblast maturation from headfold stages.

The mouse posterior primitive streak at neural plate/headfold stages (NP/HF, ~7.5 dpc-8 dpc) represents an optimal window from which hemangioblasts can be isolated. We performed immunohistochemistry on this domain using established monoclonal antibodies for proteins that affect blood and endothelial fates.

Vasculogenesis and the search for the hemangioblast.

J Hematother Stem Cell Res

April 2002

INSERM U36, Collège de France, 75005 Paris, France.

Embryonic endothelial cells (EC) are generated by two mechanisms, vasculogenesis and angiogenesis (1). The term vasculogenesis describes the de novo emergence of EC progenitors from the mesoderm, whereas angiogenesis corresponds to the generation of EC by sprouting from the pre-existing vascular network. Until recently, it was thought that vasculogenesis was restricted to the period of embryonic development, whereas in the adult, only angiogenesis contributed to EC proliferation.

ALCAM (CD166): its role in hematopoietic and endothelial development.

Blood

October 2001

Center for Tsukuba Advanced Research Alliance and Institute of Basic Medical Sciences, University of Tsukuba, Japan.

A critical role for the endothelium of yolk sac and dorsal aorta has been shown in embryonic hematopoiesis. A stromal cell line derived from yolk sac, YSCL-72, has been chosen to search for a novel molecule associated with embryonic hematopoiesis. Analysis between YSCL-72 and an adult aorta-derived endothelial cell line, EOMA, demonstrated that activated leukocyte cell adhesion molecule (ALCAM, or CD166) was specifically expressed in YSCL-72 but not in EOMA.

von Hippel-Lindau disease is a multi-system disorder characterized by hemangioblastic lesions of the central nervous system and visceral organs. Angiomatosis retinae is often the first observable manifestation of von Hippel-Lindau disease. Optometry may be the initial point of entry into the health care system for the von Hippel-Lindau patient.
