AIM:To investigate the interaction of Zot with microtubule.METHODS:Zot affinity column was applied to purify Zot-binding protein(s) from crude intestinal cell lysates. After incubation at room temperature, the column was washed and the proteins bound to the Zot affinity column were eluted by step gradient with NaCl (0.3molcenter dotL(-1) -0.5molcenter dotL(-1)). The fractions were subjected to 6.0%-15.0% (w/v) gradient SDS-PAGE and then transferred to PVDF membrane for N-terminal sequencing.Purified Zot and tau protein were blotted by using anti-Zot or anti tau antibodies. Finally, purified Zot was tested in an in vitro tubulin binding assay.RESULTS:Fractions from Zot affinity column yielded two protein bands with a M(r) of 60kU and 45kU respectively. The Nterminal sequence of the 60kU band resulted identical to beta-tubulin. Zot also crossreacts with antitau antibodies. In the in vitro tubulin binding assay, Zot coprecipitate with Mt, further suggesting that Zot possesses tubulin-binding properties.CONCLUSION:Taken together, these results suggest that Zot regulates the permeability of intestinal tight junctions by binding to intracellular Mt, with the subsequent activation of the intracellular signaling leading to the permeabilization of intercellular tight junctions.

The spurdog (Squalus acanthias Linnaeus, 1758) is a globally distributed squaliform shark that has historically been overfished but is now recovering in the northeast Atlantic. Data series on spurdog movement and habitat use have been somewhat limited to research surveys due to challenges associated with electronic tagging. Here, we offer a revised attachment method for externally attached pop-up satellite archival tags that was successful in long-term deployments on pregnant females.

In order to enrich the selection of biological ligands, realize the miniaturization analysis, and broaden the application of monolith materials for active ingredients screening and separating, we sough to construct a lipid raft @capillary monolith microcolumn affinity chromatography model. Single factor experiments and various characterization methods, including scanning electron microscopy (SEM) and thermogravimetric analysis, were employed to investigate the polymerization of the monolith column under different material ratios to determine optimal preparation conditions. Subsequently, the lipid raft from U251 cells was integrated with the monolith materials based on epoxy-based covalent crosslinking principle and characterized through SEM and immunofluorescence methods.

Contamination of heavy metals (HMs) has caused increasing concern due to their ecological toxicities and difficulties in degradation. The transport, retention, and release of HMs in porous media are highly related to their environmental fate and risk to groundwater. Column transport experiments and numerical simulations were conducted to investigate the retention and release behaviors of Cu, Pb, Cd, and Zn in the presence and absence of kaolin under varying ionic strengths and cation types.

Caltech Active Strand Cloudwater collectors are used to collect valley fog samples from Mount and North-Lebanon during 2021 for the speciation of organic matter for the first time ever. Numerous compounds including pesticides, phenols, acids, and persistent organic pollutants such as polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs), and polycyclic aromatic hydrocarbons (PAHs) have been identified in fogwater samples. They are extracted using the liquid-liquid extraction performed on the XTR chromabond columns.
