This study investigated the effect of tricyclic and atypical antidepressants on adenosine triphosphate (ATP) dependent calcium uptake by the endoplasmic reticulum of lysed synaptosomes from rat brain cortex. Tricyclic antidepressants (imipramine, desipramine, clomipramine, amitriptyline) exhibited no effect in the lower range (0.06 to 2 microM) of drug concentrations, and a concentration-dependent inhibition of calcium uptake in the upper range (6 to 200 microM). A concentration-dependent inhibition was observed for atypical antidepressants (mianserin, desmethylmianserin, venlafaxine, desmethylvenlafaxine, fluoxetine) in both the lower and the upper range of drug concentrations. Since no stimulation of calcium uptake was observed in either concentration range, it appears that the tricyclic and atypical antidepressants tested are not capable of normalizing, through their effect on the endoplasmic reticulum, an overactive calcium signal. which is possibly implicated in the etiology of affective disorders. Also, although only marginal inhibition of calcium uptake is expected at brain concentrations of tricyclics and mianserin-desmethylmianserin that are likely to be encountered during clinical use, a more substantial inhibition could occur with fluoxetine.

Regulation of mitochondrial Ca uptake is critical in cardiac adaptation to chronic stressors. Abnormalities in Ca handling, including mitochondrial uptake mechanisms, have been implicated in pathological heart hypertrophy. Enhancing mitochondrial Ca uniporter (MCU) expression has been suggested to interfere with maladaptive development of heart failure.

Primary hyperparathyroidism (PHPT) is the third most common endocrine disorder, typically caused by a single parathyroid adenoma. The diagnosis of PHPT is biochemical, and the localization of abnormal parathyroid glands is usually achieved through a combination of ultrasound and technetium-99m sestamibi (99mTc-MIBI) scans. In some cases, newer imaging modalities, such as positron emission tomography-computed tomography (PET-CT) with 18F-fluorocholine or 11C-methionine, are used as second-line methods.

Mitochondrial Ca transport regulates many neuronal functions including synaptic transmission, ATP production, gene expression and neuronal survival. The mitochondrial Ca uniporter (MCU) is the core molecular component of the mitochondrial Ca uptake complex in the inner mitochondrial membrane. MCUb is a paralog of MCU that negatively regulates mitochondrial Ca uptake in the heart and the cells of the immune system.

Despite the efficacy of bortezomib (BTZ)-based chemotherapy in treating multiple myeloma (MM) patients, chemoresistance occurs frequently over time, particularly in individuals exhibiting an initial positive response to BTZ therapy. In this study, we established BTZ-resistant MM cells and identified that suppressed expression of the hepatoma-derived growth factor (HDGF)-related protein-2 (HRP2) was a key determinant of chemoresistance in MM cells. Manipulating HRP2 expression remodeled the chemosensitivity of MM cells in vitro and in vivo.

This study examined the effects of Serendipita indica colonization and cadmium (Cd) exposure on the morphophysiological characteristics of fenugreek (Trigonella foenum-graecum). Our findings revealed that Cd stress significantly reduced plant and root height, root volume, fresh and dry weights of shoots and roots, leaf number, and the uptake of essential minerals. Cd exposure also diminished photosynthetic pigment levels while increasing certain biochemical properties and causing cadmium accumulation in plant tissues.
