Lipsitz and Goldberger proposed that there is a loss in the complexity of physiological and behavioral systems with aging and disease. Here, we show that this unidirectional view of the change in system complexity is too narrow in its consideration of the actual changes that occur with aging and disease. An increase or decrease in the complexity of a behavioral or physiological system output can occur and the direction of change is dependent on the confluence of constraints that channel the system dynamics. It is postulated that the observed increase or decrease in complexity with aging and disease is dependent on the nature of both the intrinsic dynamics of the system and the short-term change required to realize a local task demand.

Background: The ECM (extracellular matrix) provides the microenvironmental niche sensed by resident vascular smooth muscle cells (VSMCs). Aging and disease are associated with dramatic changes in ECM composition and properties; however, their impact on the VSMC phenotype remains poorly studied.
Methods: Here, we describe a novel in vitro model system that utilizes endogenous ECM to study how modifications associated with age and metabolic disease impact the VSMC phenotype.

Background: COVID-19 is an infectious disease that was declared as a pandemic and public health emergency in late 2019 and has impacted children's mental health worldwide. This study aimed to assess the general and mental health status of children during different stages of COVID-19 pandemic, and to identify the associated factors.
Methods: A cross-sectional study conducted on children aging 3 to 12 years in Kuwait during three different stages of COVID19 pandemic (pre-total curfew, during total curfew, and post-total curfew).
Introduction: Age-associated depletion in nicotinamide adenine dinucleotide (NAD+) concentrations has been implicated in metabolic, cardiovascular, and neurodegenerative disorders. Supplementation with NAD+ precursors, such as nicotinamide riboside (NR), offers a potential therapeutic avenue against neurodegenerative pathologies in aging, Alzheimer's disease, and related dementias. A crossover, double-blind, randomized placebo (PBO) controlled trial was conducted to test the safety and efficacy of 8 weeks' active treatment with NR (1 g/day) on cognition and plasma AD biomarkers in older adults with subjective cognitive decline and mild cognitive impairment.

Alpha-synuclein (ASyn), a marker of Parkinson's disease (PD) and other neurodegenerative processes, plays pivotal roles in neuronal nuclei and synapses. ASyn and its phosphorylated form at Serine 129 (p-ASyn) are involved in DNA protection and repair, processes altered in aging, neurodegeneration, and cancer. To analyze the localization of p-ASyn in skin biopsies of PD patients and melanoma.

The accumulation of ceramides and related metabolites has emerged as a pivotal mechanism contributing to the onset of age-related diseases. However, small molecule inhibitors targeting the ceramide synthesis pathway for clinical use are currently unavailable. We synthesized a safe and orally bioavailable inhibitor, termed ALT-007, targeting the rate-limiting enzyme of ceramide synthesis, serine palmitoyltransferase (SPT).
