Dual enantioselective control in asymmetric synthesis.

It is desirable and practical to produce both enantiomers of a target from the same chiral starting material by stereodifferentiation of prochiral compounds, for instance utilizing a chiral ligand derived from a natural (L) amino acid. During the past 7 years, excellent results have been achieved in several cases by multiple stereodifferentiation of chiral ligands derived from (S)-indoline-2-carboxylic acid: highly diastereo- and enantioselective pinacol coupling reactions of chiral alpha-ketoamides gave both (S,S)- and (R,R)-quaternary tartaric acid for the first time; asymmetric Diels-Alder cyclization of chiral acrylamides in the presence of Lewis acid afforded extremely high diastereoselectivities of both opposite configurations of the cyclized diastereomers depending upon the structures of chiral ligands and Lewis acids; and asymmetric alkylation of aldehydes to both enantiomers of secondary alcohols and asymmetric hydrogenation of ketones to both enantiomers of chiral secondary alcohols have been achieved using catalysts derived from (S)-indoline-2-carboxylic acid.

We here describe chemodivergent, highly enantio- and regioselective nickel-catalyzed reductive and dehydrogenative coupling reactions of alkynes, aldehydes, and silanes. A single chiral Ni-based catalyst is leveraged to directly prepare three distinct enantioenriched products (silyl-protected trisubstituted chiral allylic alcohols, oxasilacyclopentenes, and silicon-stereogenic oxasilacyclopentenes) in a single chemical operation.

Optically pure 1,2-diols and 1,3-diols are the most privileged structural motifs, widely present in natural products, pharmaceuticals and chiral auxiliaries or ligands. However, their synthesis relies on the use of toxic or expensive metal catalysts or suffer from low regioselectivity. Catalytic asymmetric synthesis of optically pure 1,n-diols from bulk chemicals in a highly stereoselective and atom-economical manner remains a formidable challenge.

Developing chiral plasmonic nanostructures represents a significant scientific challenge due to their multidisciplinary potential. Observations have revealed that the dichroic behavior of metal plasmons changes when chiral molecules are present in the system, offering promising applications in various fields such as nano-optics, asymmetric catalysis, polarization-sensitive photochemistry and molecular detection. The enantioselective [3+2] annulation of readily accessible aldimines with alkynes via C-H activation is, in principle, a straightforward and atom-efficient route for synthesizing chiral 1-aminoindenes, which are important components in a wide array of natural products, bioactive molecules, and functional materials. However, such asymmetric transformation has remained undeveloped to date due to the lack of suitable chiral catalysts. Here, we report for the first time the enantioselective [3+2] annulation of aldimines with alkynes via C-H activation using chiral half-sandwich scandium catalysts.
