Using a model of experimental leukemia in mice, we have demonstrated that tumor development depends upon interactions between preleukemic cells and their microenvironment whose functions are altered. Cytokine injections inhibit tumor development by inducing a functional restoration of this environment. Human myelodysplastic syndromes (MDS) are marrow pathologies considered as preleukemic stages. As in murine leukemias, it is possible that marrow environment could play a key role in their evolution. We currently establish a model of human hematopoiesis in NOD/SCID mice grafted with human bone fragments. We hope that this model would allow to analyse the role of the marrow stromal cells in MDS and to establish treatments restoring their functions.

Importance: Approximately one-third of patients with ERBB2 (formerly HER2 or HER2/neu)-positive (ERBB2+) metastatic breast cancer (MBC) develop brain metastasis. It is unclear whether patients with disease limited to the central nervous system (CNS) have different outcomes and causes of death compared with those with concomitant extracranial metastasis.
Objective: To assess overall survival (OS) and CNS-related mortality among patients with ERBB2+ breast cancer and a diagnosis of CNS disease by disease distribution (CNS only vs CNS plus extracranial metastasis).

Context: COVID-19 has been associated with features of a cytokine storm syndrome with some patients sharing features with the hyperinflammatory disorder, secondary hemophagocytic lymphohistiocytosis (sHLH).
Hypothesis: We hypothesized that proteins associated with sHLH from other causes will be associated with COVID-sHLH and that subjects with fatal COVID-sHLH would have defects in immune-related pathways.
Methods And Models: We identified two cohorts of adult patients presenting with COVID-19 at two tertiary care hospitals in Seattle, Washington in 2020 and 2021.

Purpose Of Review: Neuregulin 1 (NRG1) fusions are rare but actionable oncogenic drivers that occur in a variety of tumor types, including non-small cell lung cancer (NSCLC). These fusions lead to pathophysiologic activation of HER signaling pathways, promoting tumor growth, invasion, and metastasis. Current evidence suggests that NRG1 fusion-positive NSCLC does not respond well to conventional treatments such as immunotherapy and chemotherapy.
