N-hydroxysuccinimide activated Sepharose beads (HAS) were used to immobilize allotypic and idiotypic antisera for use in radioimmunoassays. These HAS antisera were compared to antisera solidified with ethylchloroformate (ECF) with respect to their reactivity, specificity, and ability to detect subspecificities of the group a allotypes. The HAS antisera rated favorably in all comparisons with ECF antisera and are far superior with respect to ease of preparation and retention of antibody activity.

Idiotypes (Ids) are unique epitopes of Ab V regions and can trigger anti-Id immune responses, but immunization with several nonadjuvanted isologous IgG mAbs has induced tolerance to their Ids. We immunized non-lupus-prone mice with 11 allotype "a" of IgG2a (IgG2a) and 4 IgG2c nonadjuvanted, isologous mAbs purified from serum-free medium. Of five IgG2a mAbs with specificity for nucleosomes, the repeating histone-DNA subunit of chromatin, four elicited an IgG1 anti-mAb response and one mAb was nonimmunogenic.

Twenty-nine different isoallotypes have been described, with 7, 4, 15, and 3 isoallotypes described for IgG1, IgG2, IgG3, and IgG4, respectively. The reactivity of anti-IgG with different isoallotypes has not been characterized.

Murine IgG responses specific for the capsular polysaccharide (pneumococcal capsular polysaccharide serotype 14; PPS14) of Streptococcus pneumoniae type 14 (Pn14), induced in response to intact Pn14 or a PPS14-protein conjugate, are both dependent on CD4(+) T cell help but appear to use marginal zone versus follicular B cells, respectively. In this study, we identify an idiotype (44.1-Id) that dominates the PPS14-specific IgG, but not IgM, responses to intact Pn14, isolated PPS14, and Group B Streptococcus (strain COH1-11) expressing capsular polysaccharide structurally identical to PPS14.
