Objective: Different facts suggest that the insulin growth factor (IGF)/ insulin growth factor-binding protein (IGFBP) system may be regulated by factors other than growth hormone. It has been proposed that, in healthy subjects, free IGF-I plays a role in glucose metabolism. The role of free IGF-I in glucose homeostasis in insulin resistance is poorly understood. This study was undertaken to evaluate the effects of acute changes in plasma glucose and insulin levels on free IGF-I and IGFBP-1 in obese and non-obese subjects.

Research Methods And Procedures: Nineteen lean and 24 obese subjects were investigated. A frequently sampled intravenous glucose tolerance test was performed. Free IGF-I and IGFBP-1 were determined at 0, 19, 22, 50, 100, and 180 minutes.

Results: Basal free IGF-I levels tended to be higher and IGFBP-1 lower in obese than in lean subjects. IGFBP-1 levels inversely correlated with basal insulin concentration. To determine the effects of insulin on the availability of free IGF-I and IGFBP-1, changes in their plasma concentrations were measured during a frequently sampled intravenous glucose tolerance test. After insulin administration, a significant suppression of free IGF-I at 22% was observed in lean subjects. In contrast, plasma-free IGF-I levels remained essentially unchanged in the obese group. The differences between both groups were statistically significant at 100 minutes (p < 0.01) and 180 minutes (p < 0.05). Serum IGFBP-1 was suppressed to a similar extent in both groups.

Discussion: These data suggest that the concentrations of free IGF-I and IGFBP-1 are differentially regulated by obesity. Obesity-related insulin resistance leads to unsuppressed free IGF-I levels.

The transition period is a crucial stage in the reproductive cycle for dams and is linked closely with postpartum recovery, reproduction performance, and health. The confronting problem in the yak industry is that transition yaks under a conventional grazing feeding regime endure nutritional deficiency since this period is in late winter and early spring of the Qinghai-Tibet Plateau with the lack of grass on natural pasture. Therefore, this study aimed to investigate the effects of perinatal nutritional supplementation and early weaning on serum biochemistry, reproductive performance, and metabolomics in transition yaks.

Article Synopsis
  • Despite the lack of a cure for osteoarthritis, researchers aimed to address mitochondrial dysfunction by developing a new treatment that uses mitochondria to deliver gene therapy via recombinant adeno-associated viral (rAAV) vectors.
  • The study demonstrated that this mitochondria/rAAV system could successfully increase the expression of insulin-like growth factor I (IGF-I) in human osteoarthritic chondrocytes, showing up to an 8.4-fold increase compared to controls.
  • The strategy not only improved cell proliferation and survival but also boosted the production of the extracellular matrix and enhanced mitochondrial function, indicating its potential as a promising treatment for osteoarthritis.

This study investigated the genetic parameters for serum IGF-I concentrations and growth traits in beef cattle. A divergent selection experiment for serum IGF-I concentration was initiated in 1989. One hundred spring-calving (50 high line and 50 low line) and 100 fall-calving (50 high line and 50 low line) black Angus cows with unknown IGF-I concentrations were randomly assigned to the two divergent selection lines.

Growth hormone (GH) is the key regulator of insulin-like growth factor I (IGF-I) generation in healthy states. However, portal insulin delivery is also an essential co-player in the regulation of the GH/IGF-I axis by affecting and regulating hepatic GH receptor synthesis, and subsequently altering hepatic GH sensitivity and IGF-I generation. Disease states of GH excess (e.

Objective: Insulin-like growth factor (IGF) is closely associated with sarcopenia, yet the causal relationship of this association remains unclear. This study aims to explore the potential causal relationship between members of the IGF family and sarcopenia from a genetic perspective through bidirectional Mendelian randomization (MR) analysis using two-sample datasets.

Methods: Five genetically predicted factors of the IGF family (IGF-1, IGF-1R, IGF-2R, IGFBP-3, IGFBP-7) as one sample, while four relevant features of sarcopenia (low hand grip strength, appendicular lean mass, whole body fat-free mass, and walking pace) as another sample, in conducting a two-sample MR analysis.
