Published: December 2001

Recombinant E. coli strains, obtained by gene engineering techniques and capable of producing human alpha-interferon and HIV proteins, were studied. The cultures under study were completely eliminated from the body of experimental animals (mice) in 48 hours, and generalization of the infectious process took place. The study revealed that these recombinant strains had low virulence and were weakly adhesive, nontoxigenic and weakly toxic. Thus, the recombinant strains under study could be classified with class 3-4 of danger according to the "Classification of Strains of Industrial Microorganisms by the Degree of Their Danger".

is an important bioresource to produce various antibacterial natural products, however, the time-consuming and labor-intensive genome editing toolkits hindered the construction and application of engineered strains, and this study aimed to establish an efficient CRISPR/Cas9n genome editing system in . Initially, the CRISPR/Cas9-mediated editing tool was employed to replace those awkward genome editing tools that relied on homologous recombination, while the off-target Cas9 exhibited high toxicity to Sf01. Therefore, the nickase mutation D10A, high-fidelity mutations including N497A, R661A, Q695A, and Q926A, and thiostrepton-induced promotor P were incorporated into the Cas9 expression cassette, which reduced its toxicity.

Combining computational modeling and experimental library screening to affinity-mature VEEV-neutralizing antibody F5.

Protein Sci

Department of Biotechnology and Bioengineering, Sandia National Laboratories, Livermore, California, USA.

Engineered monoclonal antibodies have proven to be highly effective therapeutics in recent viral outbreaks. However, despite technical advancements, an ability to rapidly adapt or increase antibody affinity and by extension, therapeutic efficacy, has yet to be fully realized. We endeavored to stand-up such a pipeline using molecular modeling combined with experimental library screening to increase the affinity of F5, a monoclonal antibody with potent neutralizing activity against Venezuelan Equine Encephalitis Virus (VEEV), to recombinant VEEV (IAB) E1E2 antigen.

Cervical cancer is predominantly caused by human papillomavirus (HPV), with oncogenic strains HPV 16 and 18 accounting for most cases worldwide. Prompt and precise identification of these high-risk HPV types is essential for enhancing patient outcomes as it enables timely intervention and management. However, the existing HPV detection techniques are time-consuming, expensive, and require highly skilled personnel.

L-valine holds wide-ranging applications in medicine, food, feed, and various industrial sectors. Escherichia coli, a pivotal strain in industrial L-valine production, features a concise fermentation period and a well-defined genetic background. This study focuses on mismatch repair genes (mutH, mutL, mutS, and recG) and genes associated with mutagenesis (dinB, rpoS, rpoD, and recA), employing a high-glucose adaptive culture in conjunction with metabolic modifications to systematically screen for superior phenotypes.
