[Use of fibrin-collagen plates in abdominal surgery].

The article presents results of experimental substantiation and clinical use of fibrin-collagen plates Tachocomb. They were used in clinical practice in 28 patients with different bleedings under conditions of open and little invasive surgery. The preparation not only increases the mechanical strength of the surgical suture, but also has a stimulating effect on reparative processes. The author proposes to use Tachocomb for strengthening the sutures and anastomoses in peritonitis, intestinal obstruction and interventions in the area of the tissues altered due to infiltration.

In an effort to improve the long-term patency of vascular prostheses several groups now advocate seeding autologous endothelial cells (ECs) onto the lumen of the vessel prior to implantation, a procedure that involves pre-treating the prosthesis material with fibrin, collagen and/or other matrix molecules to promote cell attachment and retention. In this study, we examined the degree to which human umbilical venous endothelial cells (HUVECs) adhered to three materials commonly used polymeric vascular prosthesis that had been coated with the same commercial extra cellular matrix proteins, and after exposure to fluid shear stresses representative of femoro-distal bypass in a cone-and-plate shearing device. We quantified cell number, area of coverage and degree of cell spreading using image analysis techniques.

Experiments in 5 pigs and 18 mongrel dogs have shown the possibility to close perforating ulcers with plates of biopolymer "Tachocomb". The method is proposed as an alternative to suturing perforations in the zone of pylorus in order to prevent stenosis of the pyloric part of the stomach. The experimental data obtained have confirmed good adhesive and stimulating properties of the fibrin-collagen substance.

In this paper we describe a new and simple technique for studying cell movement in vitro. This method is based upon the migration of endothelial cells on polystyrene bead monolayers. It permits the rapid preparation and screening of multiple 96-well plates for effects on migration, requires fewer samples per test and has the potential for precise quantitation by digital image analysis.
