DNA-binding activity and cytotoxicity of the extended diphenylfuran bisamidines in breast cancer MCF-7 cells.

The DNA binding properties of three novel extended diphenylfuran bisamidines (1-3) possessing different dicationic terminal side chains were studied. The ultrafiltration assay showed that bisamidines 1-3 have significant affinity for DNA. The DNA-binding data for bisamidines 1-3 using homopolymers poly(dA-dT)- poly(dA-dT) and poly(dG-dC)- poly(dG-dC), indicated that these compounds show moderate specificity for AT base pairs. We studied the cytotoxicity effects of bisamidines 1-3, Hoechst 33258 and DAPI (4',6-diamidino-2-phenylindole) in cultured breast cancer MCF-7 cells. The bisamidines 1-3 showed comparable antitumour activity to Hoechst 33258, but were substantially more cytotoxic compared to DAPI. These data show that in broad terms the cytotoxic potency of bisamidines 1-3 in cultured breast cancer MCF-7 cells decreases with the size of the alkyl group substituent (cyclopropyl>isopropyl>cyclopentyl), in accord with their increases in DNA affinity, as shown by the binding constant values.

We report the iridium-catalyzed, stereoselective conversion of secondary alcohols or ketones to -1,3-diols by the silylation of secondary C-H bonds γ to oxygen and oxidation of the resulting oxasilolane. The silylation of secondary C-H bonds in secondary silyl ethers derived from alcohols or ketones is enabled by a catalyst formed from a simple bisamidine ligand. The silylation occurs with high selectivity at a secondary C-H bond γ to oxygen over distal primary or proximal secondary C-H bonds.

A Zn complex of a C -chiral bisamidine-type sp N bidentate ligand (L ) possessing two dioxolane rings at both ends catalyzes a highly efficient quinone asymmetric Diels-Alder reaction (qADA) between o-alkoxy-p-benzoquinones and 1-alkoxy-1,3-butadienes to construct highly functionalized chiral cis-decalins, proceeding in up to a >99:1 enantiomer ratio with a high generality in the presence of H O (H O:Zn =4-6:1). Ureas of chiral diamines are prominent features of therapeutics, chiral auxiliaries, and intermediates in complex molecule synthesis. Although many methods for diamine synthesis are available, metal-free enantioselective alkene functionalizations to make protected 1,2- and 1,3-diamines from simple achiral starting materials are rare, and a single reagent that accesses a cross-section of each congener with high enantiomeric excess is not available. We describe a method to synthesize enantioenriched cyclic 5- and 6-membered ureas from allylic amines and an isocyanate using a C2-symmetric BisAmidine (BAM) catalyst that delivers N-selectivity from an ambident sulfonyl imide intermediate, overcoming electronic and steric deactivation at nitrogen.
