The triad of infection--the dynamic equilibrium between pathogen, host, and drug--offers both opportunities and challenges for effectively delivering the right amount of drug to the right place for the right amount of time. The delivery system and drug must cooperate to achieve delivery and substantivity at the target site. Given the amount of variability introduced by the dosage form, it takes plenty of trial and error to get the right drug in the right vehicle so it can reach the targeted tissue. The deeper the target tissue, the greater the complexity of drug delivery and effect.

Background: Doxepin (DX) is used orally to relieve itching but can cause side effects like blurred vision, dry mouth, and drowsiness due to its antimuscarinic effect. To reduce these adverse effects and improve skin permeation, DX is being developed in topical formulations. This study aims to improve DX skin absorption by developing a microemulsion (ME) formulation (ME-DX).

Cyclosporin A (CsA) is a drug used to prevent immune rejection in corneal transplantation. Most grafts performed today are endothelial grafts which are complicated with poor penetration of CsA into the endothelium due to its hydrophobicity. To improve CsA penetration into the corneal a new ocular formulation of CsA 2% with Miglyol was developed and is commercially available.

The use of cannabis-related products is currently experiencing extraordinary growth in popularity in the European and US markets. A wide variety of cannabis-related products have emerged, including oils, tinctures, edibles, topicals, cosmetics, and even beverages and sweets, offering the purported medical benefits without the psychoactive effects associated with Cannabis sativa. However, there is a significant gap in our understanding of bioaccumulation processes and their long-term effects, particularly as cannabinoids are highly lipophilic molecules.

The present work focuses on the production of sildenafil co-evaporates loaded emulgels as topical dosage forms for the treatment of premature ejaculation and erectile dysfunction. Topical administration of sildenafil citrate (SILD) co-evaporates is expected to improve the bioavailability profile of the drug and to avoid the severe side effects accompanying the traditional SILD dosage forms, especially for prohibited cardiovascular cases. Firstly, the solubility of SILD was improved via solid dispersion via co-evaporation technique using PEG-5KDa and PVP-K90 as hydrophilic carriers.

Skin cancer stands as a challenging global health concern, necessitating innovative approaches to cure deficiencies within traditional therapeutic modalities. While conventional drug delivery methods through injection or oral administration have long prevailed, the emergence of topical drug administration presents a compelling alternative. The skin, aside from offering a swift and painless procedure, serves as a reservoir, maintaining drug efficacy over extended durations.
