Purpose: A significant correlation has been found between p53 mutation and response to chemotherapy or radiotherapy. To determine the prognostic value of p53 mutation in patients with locally advanced esophageal carcinoma treated with definitive chemoradiotherapy, p53 mutation was analyzed using the biopsied specimens taken for diagnosis.
Methods: Concurrent chemoradiotherapy was performed for 40 patients with severe dysphagia caused by esophageal squamous cell carcinoma associated with T3 or T4 disease. Chemotherapy consisted of protracted infusion of 5-fluorouracil, combined with an infusion of cisplatinum. Radiation treatment of the mediastinum was administered concomitantly with chemotherapy. The p53 gene mutation was detected by fluorescence-based polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) methods. DNA sequences were determined for DNA fragments with shifted peaks by SSCP methods.
Results: Of the 40 patients, 15 had T3 disease and 25 had T4 disease; 11 patients had M1 lymph node (LYM) disease. Of the 40 patients, 13 (33%) achieved a complete response. The median survival time was 14 months, and the 2-year survival rate was 20%. Among the 40 tumor samples, p53 mutation was detected in 24 tumors (60%). The survival rate in the 24 patients with p53 mutation did not differ significantly from that in the 16 patients without p53 mutation. In contrast, the 15 patients with T3 disease survived longer than the 25 patients with T4 disease (P = 0.016); however, the survival rate in the 11 patients with M1 LYM disease did not differ significantly from that in the 29 patients without M1 LYM disease.
Conclusion: Concurrent chemoradiotherapy is potentially curative for locally advanced esophageal carcinoma, but p53 genetic abnormality has no impact on prognosis.

Background: The clinical implications of different EML4-ALK fusion variants remain poorly elucidated in the era of second-generation ALK inhibitors.
Methods: This was a retrospective cohort study, wherein patients diagnosed with locally advanced or metastatic non-small cell lung cancer harboring EML4-ALK fusion were stratified into two cohorts based on their first-line treatment: Cohort 1 received alectinib, while Cohort 2 received crizotinib. Statistical analysis was employed to investigate the impact of different EML4-ALK variants and TP53 status on the efficacy of first-line ALK-TKIs.

The role of p53 expression in colorectal cancer (CRC) was investigated in this immunohistochemical analysis of 110 CRC patients. The study aimed to explore the relationship between p53 expression and clinicopathological features, such as tumor grade, size, lymph node involvement, and molecular subtypes. The mean age of patients was 52.

Introduction: 5-aminolevulinic acid (5-ALA) fluorescence used in glioma surgery has different intensities within tumors and among different patients, some molecular and external factors have been implicated, but there is no clear evidence analyzing the difference of fluorescence according to glioma molecular characteristics. This study aimed to compare molecular factors of glioma samples with fluorescence intensity to identify potential cofounders and associations with clinically relevant tumor features.
Methods: Tumor samples of high-grade glioma patients operated using 5-ALA for guided resection were included for comparative analysis of fluorescence intensity and molecular features.

Rationale: Follicular tumors with uncertain malignant potential of the thyroid are a new classification of thyroid tumors in the World Health Organization (2017). Only a few cases of evaluating the value of ultrasound combined with pathological examination in diagnosing follicular tumors with uncertain malignant potential of the thyroid have been reported in recent decades.
Patient Concerns: A retrospective analysis was performed on 18 patients with follicular tumors with uncertain thyroid malignant potential who underwent preoperative ultrasonography and were confirmed by operation and pathology in our hospital from January 2019 to November 2024.

Introduction: Prostate cancer guidelines recommend molecular analysis of biomaterial following resistance to first-line systemic therapy in order to identify druggable mutations. We report on our results of molecular analysis of tissue specimens via next generation sequencing (NGS) in men with metastatic castration resistant prostate cancer (mCRPC).
Patients And Methods: In all, 311 mCRPC patients underwent NGS analysis from biopsy samples of progressive metastatic lesions or archival radical prostatectomy specimens.
